Radix Polygoni Multiflori,the dried roots of Polygonum multiflorum Thunb.(Polygonaceae)has been used widely as a traditional Chinese medicine for a long time.It has been used for its anti-aging,anti-inflammatory,antioxidant,and hair-blacking properties.In this thesis,the chemical constituents of Radix Polygoni Multiflori were extracted,isolated and identified,and pharmacological activities of these constituents were also evaluated.Twenty-six compounds were isolated from Polygonum multiflorum by organic solvent extraction,silica gel column chromatography,reversed gel column chromatography,Sephadex LH-20 column chromatography,and semipreparative HPLC,including nine quinones(1-9),six anthraquinones(11-16),four flavonoids(17-20),and four amides(21-24).The structures of these compounds,including physcion(1),emodin-8-O-?-D-glucopyranose(2),emodin(3),fallacinol(4),1,5-dihydroxy-3-methoxy-7-methyl-anthraquinone(5),emodin-8-P-D-(6'-O-acetyl)glucoside(6),polynimitin B(7),2-methoxy-6-acetyl-7-methyljuglone(8),6-methoxyl-2-acetyl-3-methyl-1,4-naphthoquinone-8-O-?-D-glucopyranoside(9),torachrysone-8-O-?-D-glucoside(10),(E)-2,3,5,4'-Tetrahydroxy-stilbene-2-O-?-D-glucopyranoside(11),(Z)-2,3,5,4'-Tetrahydroxy-stilbene-2-O-?-D-glucopyranoside(12),(E)-2,3,5,4'-tetrahydroxystilbene-2-O-?-D-glucopyranosyl-5-O-?-D-glucopyranoside(13),(E)-2,3,5,4'-tetrahydroxystilbene-2-O-(4"-O-?-D-glucopyranosyl)-?-D-glucopyranoside(14),(E)-2,3,5,4 '-tetrahy droxy stilbene-2-0-?-D-glucopyranosyl-4'-O-?-D-glucopyranoside(15),polynimitin C(16),vicenin-2(17),tricin(18),naringenin(19),(-)-catechin(20),grossamide(21),n-trans-feruloyltyramine(22),cannabisin D(23),cannabisin E(24),2,5-dimethyl-7-hydroxychromone(25),and p-formaldehyde(26),were identified by their NMR and MS spectra.By using the model of LPS-induced RAW264.7 cells,NO inhibition rate and TNF-a inhibition rate were investigated.The results indicated that four anthraquinones,one anthraquinone,three flavonoids,and four acid amides showed high anti-inflammatoryactivity.They are emodin-8-O-?-D-glucopyranose(2),2-methoxy-6-acetyl-7-methyljuglone(8),torachrysone-8-O-?-D-glucoside(10),(E)-2,3,5,4'-tetrahydroxystilbene-2-O-?-D-glucopyranosyl-4'-O-?-D-glucopyranoside(15),tricin(18),naringenin(19),(-)-catechin(20),grossamide(21),n-trans-feruloyltyramine(22),cannabisin D(23),and cannabisin E(24).The NO inhibition rates are range from 36%to 126%(50 ?M),and the TNF-a inhibition rates are range from 72.68%to 99.84%(50 ?M).Emodin-8-O-?-D-glucopyranose(2),2-methoxy-6-acetyl-7-methyljuglone(8),tricin(18),naringenin(19),(-)-catechin(20),grossamide(21),and n-trans-feruloyltyreamine(22)showed high NO inhibition rates of 120%,126%,72%,82%,88%,90%,and 88%,respectively.2-Methoxy-6-acetyl-7-methyljuglone(8),(E)-2,3,5,4'-tetrahydroxystilbene-2-O-?-D-glucopyranosyl-4'-O-a-D-glucopyranoside(15),naringenin(19),and grossamide(21)showed high TNF-a inhibition rates of 97.84%,97.85%,99.84%,96.52%,respectively.
