The Solid Phase Extraction Column(SPE column)is a kind of sample pretreatment device used for extraction,separation and enrichment,which developed by combination of liquid-solid extraction and chromatography.At present,based on the lack of safety management in Chinese food market,food problems are constantly emerging.In order to quickly and efficiently perform food safety testing,the combination of solid phase extraction and chromatography has been extensively applied in the detection of various food,environmental samples,and target samples of agricultural and livestock products.This article is divided into three chapters and the research content is as follows:Chapter one: Overview of the current status and measurement metheod of preservatives and antibiotics,and the background and content of the topics selection.Chapter two: I established a solid phase extraction-gas chromatography method for the determination of five P-hydroxybenzoate preservatives in vegetable oils.The optimization of conditions such as stationary phase,column temperature on gas phase separation conditions,sample extraction and other factors were investigated,and the best conditions for the separation of ester preservatives from the sample were finally obtained.In the linear response range of 0.500-100 mg/L,there was a good linear relationship between the five ester preservatives and the correlation coefficient(R)was 0.9993-0.9996.The limit of detection was 0.20~0.22 mg/L,the recovery was 76.88~115.2%,and the relative standard deviation was 2.3~3.9%(RSD,n=5).This method is of reference for the detection of ester antiseptics in edible oils in the future.Chapter three: I established a solid phase extraction-high performance liquid chromatography method for the determination of sulfonamides in vegetables.The separation factors of standard samples such as mobile phase and the optimization of sample extraction conditions were studied.In the linear response range of 1.00 to 100 mg/L,there was a good linear relationship among the eight sulfonamides.The correlation coefficient(R)was 0.9997-0.9999.The limit of detection was 0.025-0.075 mg/L,the recovery was 68.92-122.7%,and the relative standard deviation was 1.8-4.5%(RSD,n=5).This method is useful for the detection of possible residual sulfa antibiotics in vegetables.
