The Filamentous actinomycetes of gram positive is Streptomyces,produced multiple types of the secondary metabolites that have important vality,such as antibiotics,immunomodulators and so on.Streptomyces produce synthetic secondary metabolites that have a variety of water-soluble or fat-soluble pigments,withing high industrial value.Natural pigments have the best factor is high safety,has been caused by various areas concern.The natural pigment of Streptomyces secondary metabolites is not only be used as dyes,but also have antibacterial effect,and other advantages.The advantage of double effct have great prospects for development.The corn starch and soybean meal can provide carbon and nitrogen source for Streptomyces,The ability of produce pigment are higher than other industry medium is streptomycin,Add inorganic salts to medium have no significant effect for Streptomyces pigment.Streptomyces fermentation by liquid shake method,getting on Streptomyces fermentation,The most significant of produce pigment at 7-8 d for Streptomyces,The mycelia color more deeper.The raw material choosed Streptomyces Fermentation mycelia at seventh day,by Box-Behnken experimental design,use Response surface method that determine extraction process of optimize the pigment,We determine the optimum extraction conditions:(1)the conditions of extracting orange-red pigment,the extraction of methanol,the solid and liquid ratio was 50:1,the ultrasonic power was 100 w,the extraction time was 30 min.(2)The conditions of extracting orange red pigment,methanol extraction,the ratio of solid and liquid was 50:1,ultrasonic power was 100 W,extraction time was 15 min.Study on the TLC and silica gel column layer of Streptomyces pigment extracts were isolated and identified column elution conditions:elution conditions of silica gel column for ethyl acetate hexane(4:1,V/V)was selected with silica gel column can separate the 3 components.Study of Streptomyces gold red,orange red,orange and yellow 4 kinds of pigment distribution in different hexane-ethyl acetate-methanol-water system in proportion,found that the orange and orange in phase K values were similar,but the red and yellow in the upper and lower phase difference was determined.K hexane:ethyl acetate:methanol:water =6:4:7:3 is determined from Streptomyces four kinds of optimal conditions for separation.The experiment proves that compared Gao 1 medium,Streptomyces with soybean meal powder and maize starch as carbon and nitrogen source,the secondary metabolites production of pigment is relatively high,adding inorganic salt no significant increase of secondary metabolites.Finally,by ethanol on pigment dyeing of wool Streptomyces to explore the fabric process:water 1:4,bath ratio 1:30,dyeing temperature 95 ?,dyeing time 80min,pigment content 5%o.w.f.,without additives,not the pH value is adjusted to the optimum conditions.
