Objective:To investigate the effect of the composite probiotics milk on the db/db diabetes mice.The Compound probiotics contain five kinds of probiotics such as Lactobacillus kefiranofaciens,Lactobacillus plantarum,Lactobacillus helveticus,Lactococcus lactis and Issatchenkia orientalis.Methods:1.In accordance with the number of viable cells examined,pH,and other indicators,prepared in the laboratory complex probiotic camel milk.2.40 db?db mice diabetic mice were divided into four groups and treated for 6 weeks with Metformin HCL(0.3g·kg-1)or composite probiotics milk?4×108 lactic acid bacteria+1×106 Issatchenkia orientalis?CFU?(4×1010 lactic acid bacteria+1×108 Issatchenkia orientalis)CFU and a diabetic control group.In addition,10 C57BL/Ks mice as normal control group were used for comparison.Fasting blood glucose?FBG?and body weight?BW?were measured before treatment and every week thereafter.All mice received OGTT in the end of 6weeks to evaluate it glueose metabolism.Glycated hemoglobin?HbAlc?,C-peptide?CP?,triglycerides?TG?,Total cholesterol?TC?,low-density lipoprotein cholesterol?LDL-c?,high density lipoprotein cholesterol?HDL-c?,24 hour urinary microalbumin?24 hmalb?,urine ketone?KET?,urine sugar pancreas and liver tissue were tested after 6 weeks.3.The interactions of lactic acid bacterium and Issatchenkia orientalis.Results:High dose composite probiotics milk decreased FBG,OGTT,HbAlc and IRI increased C-peptide?P<0.05?levels,decreased serum TC,TG and LDL-c?P<0.001?,increased HDL-c levels,improved the liver and kidney function compared to diabetic controls.Histological analysis showed composite probiotics milk protected the function of islets and liver of db/db mice diabetic mice In details,the growth of Lactobacillus kefiranofaciens and Lactococcus lactis?P<0.05?was advanced by the addition of Issatchenkia orientalis;Issatchenkia orientalis had no obvious effect on the growth of Lactobacillus helveticus?P>0.05?during the fermentation process.At the same time,the addition of lactic acid bacteria could inhibit the growth of Issatchenkia orientalis?P<0.05?.In addition,a positive interaction between lactic acid bacteria and Issatchenkia orientalis was observed what could keep the viability of each other during the cold storage.Conclusion:The results of this study indicates that composite probiotics milk decreased FBG,OGTT and HbAlc,increased C-peptide,modulated lipid metabolism and protected liver and kidney injury in db?db mice diabetic mice,which may be related to protecting the function of islets.
