The Synthesis And Research Of Diclazuril And Its Intermediate

Diclazuril which belongs to triazine phenylacetonitrile compound is a novel chemistry synthesize anticoccidial drug.Diclazuril has been widely used in chicken coccidiosis because of its broad anticoccidial spectrum,good efficacy,fast metabolism in chicken,low toxicity,no residue.In this thesis,the traditional production process of diclazuril was optimized and a new synthetic route of diclazuril was proposed;intermediate in new synthetic route was researched.This thesis was divided into three chapter.The first chapter gived a brief introduction about coccidiosis and anticoccidial drug,presented an inverse synthetic analysis of diclazuril,outlined the synthetic route of diclazuril and summarized some decarboxylation reaction of heterocyclic compounds,hydrazine derivatives,1,2,4-triazole-5-one derivatives and 1,2,5-triazin-4,6-dione derivatives.The second chapter was experimental content:2-(4-chlorophenyl)-2-(2,6-dichloro-4-(3,5-dioxo-4,5-dihydro-1,2,4-triazin-2(3H)-yl)phenyl)acetonitrile can be synthesized from 2-(3,5-dichloro-4-((4-chlorophenyl)(cyano)methyl)phenyl)-3,5-dioxo-2,3,4,5-tetrahydro-1,2,4-triazine-6-carboxylic acid using thiourea as more efficient catalyst.The optimum process conditions which its yield was 77% and content of product reached more than 97.5% had been determined through effects of the reaction solvent,temperature,feed ratio on the reaction.The third chapter studyed a new routes synthesis of diclazuril.Diclazuril can be synthesized from 4-amino-2,6-dichloro-?-(4-chlorophenyl)benzyl cyanide by hydrazine,hydrazone,cyclization,ring expansion reaction.At present,1,2,4-triazole-5-one derivatives was synthesized and its structure was confirmed,the optimum reaction conditions were investigated and determined.
