Oolong tea is a Chinese endemic tea,Oolong tea processing plays an important role in the six kinds of tea processing.Fine manipulation is a unique processes in Oolong tea processing,it is a key to shape,smell,color,taste,bottom of Oolong tea during fine manipulation with repeatedly replaced of shaking and drying indoors.Simultaneously,the physicochemical changes of leaves is complex and rich in the fine manipulation processes,it has been a hot topic in the industry.In this paper,it researched the change of lipid peroxidation and protective enzyme of leaf mitochondria under different temperature during fine manipulation,which exists a significant academic value for further enrich the research of theory and technology in Oolong tea processing.In this paper,the test material is Camellia.sinensis cv.Mingke 1,and another name is Camellia.sinensis cv.Jinguanyin,the experimental factors were different fine manipulation temperature(T)and treatment of green leaves(A).There were three temperature gradient-low(T1),medium(T2)and high temperature(T3)and different treatment of green leaves-shaking(A1)and standing(A2),six treatments totally.It was designed four times shaking by manual in air conditioner room.In the paper,it was researched the changes of water content,lipid peroxidation and protective enzyme and correlation of all the above indexes.1.The water content of leaves was decreased on the whole,but the extent had its own characteristic.The water content change is slow in the whole process of fine manipulation,and the change range is stable under A2 treatment.The water content change has a difference between prior period and later period in the whole process of fine manipulation under A1 treatment.The water content respectively decreased by 5.83%,8.26%,5.34%among low,medium and high temperature during fine manipulation later period,and decreased by 4.17%,1.61%,3.49%during fine manipulation prior period,the later period water content change was slower than the prior period.Meanwhile,the decrease speed of water content was faster and the change range was larger in the prior period,and it was slower and smaller in the later period.Water content,lipid peroxidation and protective enzyme were negative correlations at different levels under A1 and AZ treatment,which showed the content and activity of lipid peroxidation and protective enzyme were improved in different extent with the water content of leaves decreased.2.1t had a significant difference of mitochondrial MDA content,POD activity,CAT activity in different fine manipulation temperature(P<0.05),and it had no significant difference between low and medium temperature,and the mitochondrial MDA content,POD activity of high temperature was significantly higher than low and medium temperature(P<0.05),the mitochondrial CAT activity of high temperature was significantly higher than medium temperature(P<0.05).The significant difference of mitochondrial H2O2 content and SOD activity were not obvious among different fine manipulation temperature.3.There were significant differences in different treatment of green leaves(P<0.05),and there existed a extremely significant difference in different treatment of green leaves(P<0.01).There was no significant difference of MDA content and POD activity in different treatment of green leaves,but the change of MDA content for A1 treatment was richer than A2 treatment.And the range of variation of MDA content respectively were 0.16-2.77,0.28-5.19,1.19-6.25 nmol/mg under A1 treatment,these were 0.43-2.62,0.44-4.09,1.05-2.35 nmol/mg under A2 treatment,which showed the range of variation also was higher.The accumulation of POD activity respectively were 0.99,2.03,3.65 AOD470/mg Pro-min among low,medium and high temperature during fine manipulation later period under A1 treatment,and 0.67,0.73,2.08 AOD470/mg Pro-min under A2 treatment.It indicated that the accumulation of POD activity under A1 treatment is higher than A2 treatment in the later period.4.The mitochondrial MDA content and CAT activity in interaction of fine manipulation temperature and different treatment of green leaves also existed significant differences(P<0.05).And there were different between significant difference and extremely significant difference about mitochondrial H2O2 content,MDA content,SOD activity,POD activity and CAT activity of each inter-process.5.Lipid peroxidation was related to protective enzyme during fine manipulation.Under A?treatment,there were extremely significant positive correlations between H2O2 content,MDA content,SOD activity,POD activity and CAT activity(r=0.645,r=0.780,r=0.865,r=0.843,P<0.01),and SOD activity and POD activity had a significant positive correlation(r=0.543,P<0.05),the SOD activity,POD activity and CAT activity had extremely significant positive correlations(r=0.708,r=0.635,P<0.01).The H2O2 content and CAT activity had an extremely significant positive correlation(r=0.597,P<0.01)under A2 treatment.It indicated that the correlations among all indexes of A1 treatment was richer than A2 treatment,and these of A1 treatment between the various indexes was closer.Simultaneously,protective enzyme played a positive and crucial role in eliminating ROS,and each index of protective enzyme existed a significant cooperative effect.
