OBJECTIVE1 Investigated the feasibility of bile salt and lecithin as mixed surfactant to preparate microemulsion containing cholesterol,and optimized the microemulsion formulation.2 Investigation the content glycyrrhizin and baicalin of Longqincao compound extract,select the best extraction method.3 Extraction process for Longqincao compound microemulsion extraction efficiency to determine the optimal extraction process.METHODS1 The microemulsion was optimized by mixture experiment methods.The independent variables selected were surfactant,co-surfactant and oil;dependent variables(responses)were particle size.Mathematical equations and response surface plots were used to relate the dependent and independent variables.Optimized formulation factors were selected by feasibility and grid search,with Malvern grain trail determination instrument determination grain trail,transmission electric mirror directly observation,and studied the stability of the microemulsion.2 Using percolation method,cold soak method,ultrasonic method,water bath method to extract Longqincao compound,the extracting solution was measured glycyrrhizin,baicalin content by HPLC.3 Through orthogonal test method,inspect immersion time,particle size of material,the solid-liquid ratio,the velocity of percolation effect on the extraction rate,HPLC method was used to determine the index of content,and carries on thestatistical analysis.RESULTS1 Optimization of microemulsion with average particle size of 19.12 nm,the polydispersity index(PDI)averaged 0.153,under transmission electron microscopy with round shape,uniform particle size distribution,good physical stability,under high speed centrifugation,high temperature,room temperature,low temperature were stable.2 The highest extraction rate of percolation method.In extraction of ethanol,contents of liquiritin was 5.16mg/ml,the content of baicalin as the 94.73mg/ml;in extraction of microemulsion,contents of liquiritin was 5.28mg/ml,the content of baicalin as the 122.43mg/ml.3 According to the analysis results,the optimal extraction process is as follows:37? impregnated 36h,medicinal powder to 20 mesh,material liquid ratio 1:8,the velocity of percolation 3-5ml/min.Biocompatible microemulsion formulations using the best extraction technology of compound extracted Longqincao extract:glycyrrhizin content of 80mg,baicalin as 604.66mg,total protein content as56.88mg.Conclusion:1 A lecithin,cholesterol,bile salts,for the preparation of microemulsion formulations are biocompatible feasible.2 Formulation mixed microemulsion method for screening is feasible to screen the composition formula:S:COS:Oil:H2O=0.232:0.031:0.013:0.724..3 Follow screened prescription form stable microemulsion formulation biocompatible nature of the preparation,particle size between 10-100nm,meet the definition of microemulsion.4 Best extraction method uses a biocompatible microemulsion formulation extracted Longqincao compound is:37? constant temperature percolation;optimum extraction of:37? dipping 36h,herbs crushed to 20 mesh,solid-liquid ratio 1:8,percolation rate 3-5ml/min.
