AimTo elucidate the chemical components in GHB and the represent of reducing asthenia heat prescription Da Bu Yin Pill et al.and their absorbed components or metabolites in rat plasma after oral administration of GHB and the prescription of DBYW et al.by using UPLC-MS/MS and MassLynx data processing technology,provide a model for analyzing the constituents of Chinese medical formula in vitro and in vivo and help to explore the effective substance of GHB?DBYW?LBZSW?ZSW according to Serum Pharmacochemistry of TCM.In this paper,the correlation and function orientation of GHB was revealed.At last,the hypothesis of "the efficacy of traditional Chinese medicine taking on its corresponding material basis" was verified.MethodsAccording to the theory of Serum Pharmacochemistry of TCM,UPLC-MS/MS technique with the MS Scan acquisition mode was applied to detect and characterize the constituents of GHB and the prescription of DBYW et al.and the components and their metabolites absorbed in rat plasma after oral administration of GHB and the prescription of DBYW et al.The UPLC-MS/MS chromatograms acquired were processed through the intuitionistic comparation and the MassLynx data processing technology respectively.Results1 Analysis of the constituents in vitroAn injection run of the GHB and the prescription of DBYW et al.sample was finished in 24 min,and excellent chromatographic separation and sensitive detection were obtained.By elucidation of mass spectra at both high and low collision energies,56 compounds were detected in GHB,91 compounds were detected in DBYW,87 compounds were detected in LBZSW,87 compounds were detected in ZSW.2 Analysis of the constituents in vivoBased on the developed method of fingerprint analysis of the GHB and the prescription of DBYW et al.an injection run of the sample of dosed rat plasma was finished in24.0 min.Through the intuitionist comparisons of the three chromatograms and applications of MassLynx data processing technology,29 compounds derived from GHB(salt);39 compounds derived from DBYW;35 compounds derived from LBZSW;37 compounds derived from ZSW;ConclusionAccording to Serum Pharmacochemistry of TCM,UPLC-MS/MS was used to establish excellent methods for analyzing the constituents of GHB and the prescription of DBYW et al.in vitro and in vivo.The complete compounds of GHB and the prescription of DBYW et al.in vitro and in vivo were explored and charectered by MS Scan and Masslynx data processing technology.The compounds found in dosed serum could be the effective Substances of GHB and the prescription of DBYW et al.
