The fields of mangrove endophytic fungus and aquatic fungus have become a hotpot for research in the world.Numerous compounds with strong activity and novel skeletons were isolated from mangrove endophytic fungus and aquatic fungus.It plays an important role in many fields such as immunomodulatory angents,anti-tumor,anti-oxidation and the cure of hypertension.The study on the secondary metabolites of mangrove endophytic fungi and aquatic fungi lays theoretical basis for developing lead compounds.Ten species of mangrove plants were collected from the estuary of Beilun Guangxi,including Hibiscus tiliaceus,Bruguiera gymnorrhiza,Aegiceras corniculatum,Excoecaria agallocha,Acanthus ilicifolius,Cerbera manghas,Kandelia candel,Clerodendrum inerme,Avicennia marina and Acrostichumaureum.Fourty endophytic fungi were isolated from fresh leaves and stems by strict aseptic operation.Three fungal strains(efKcY2041,efAiJ2043,efHtY2041)were incubated on solid state fermentation for 40 days at room temperature.The thalus were soaked in methanol three times and filtered through cheesecloth.The soaking liquid was concentrated below 50?by rotary evaporator to get the crude extract.Twenty compounds were isolated by column chromatography,recrystallization and Sephadex LH-20 on silica gel.By the way of MS and NMR,their structures were identified as stigmasterol,ergosterol-7,22(E)-diene-3?,5?,6?-triol,chrysophanol,mannitol,succinic acid,oxalic acid,myristic acid,12-hydroxy stearic acid,stearic acid,indole-3-acetic acid,protocatechuic acid,p-hydroxybenzoic acid,benzoic acid,cinnamic acid,salicytic acid,2-methy-6-hydroxy benzoic acid,4-hydroxybenzoic acid ethyl ester,hydroquione,p-hydroxybenzaldehyde and vanillin.Two aquatic fungus(LC1701,LC1715)were incubated on solid state fermentation for 40 days at room temperature.The crude extract was extracted with ethyl acetate.Fourteen compounds were isolated using column chromatography,recrystallization and Sephadex LH-20 on silica gel.By the methods of MS and NMR,their structures were identified as emodin,?-sitosterol,cholesterol,methyl 4-hydroxybenzoate,dimethyl terephthalate,methyl 3-hydroxy-4-methoxybenzoate,p-hydroxybenzene propanoic acid,p-methoxyphenylacetic acid,nicotinic acid,4-hydroxybenzeneacetamide,acetanilide,4-methoxyphenol,4-hydroxyphenethyl alcohol and acetovanillone.Eighteen active compounds were choosed for virtual screening.Seven targets related to compounds were NOS,SAMDC,GSTs,MIF,ODC,MAPK and CYP450.These enzymes have imporant application in anti-inflammatory,anti-tumor,cell differentiation,detoxification,immune disease and drug metabolism.These results provide theoretical guidance for explaning mechanism of active pharmacologically compounds and develop marine drugs.
