Galangal,one of the most famous China traditional medicine,has been widely used in pharmaceutical,food,cosmetics and reservation of fruit and vegetable,with a great quantity of functional components.So far,chemistry producing,taking essential oil of galangal as the primary product,only gains the oil through the traditional methods.In addition,many ingredients,as usual,would leave in the residue of galangal,which have no practice to be used,cause the waste of functional elements from galangal.This circumstance would change if the extraction method get improved through study and will be able to gain more than one functional component.To get a new way to develop galangal industry,we take galangal as raw material and find a new method to extract essential oil and flavone from galangal.Firstly,we optimize the essential oil extraction and evaluate the quantity and quality of flavone from the residue,aiming to gain essential oil as much as possible and keep high quantity and high quality of flavone from residue.Then,we optimize the flavone extraction to improve the yield.Last but not least,after purification,the flavone could enhance purity and antioxidant activity,which mean we gain a new practice to extract essential oil and flavone from galangal at one continuously course.Here is the research:(1)Study on essential oil extractionTaking galangal as raw material,evaluating yield and extraction time,we assess the influence of shape and size of sample,solid-liquid ratio,soak time on essential oil extraction and residue.As the result,the optimized conditions:powder size smaller than0.850 mm,solid-liquid ratio of 1:12,soak time of 30 min were obtained,and at these conditions,the yield of essential oil is 0.82 mL in 45 min,with little effect on quantity and quality of flavone from residue.Besides,we establish kinetic model based on the theory of essential oil extraction,which consists of surface washing and internal diffusion,to explain the result of extraction.In addition,the composition of galangal essential oil was identified,and we found the mean constituents were 1,8-Oxido-p-menthane(40.22%),Camphene(6.67%),?-Pinene(7.96%),?-Pinene(5.76%),Terpinol(4.34%)and their like,most of which were Terpenes.(2)Study on flavone extractionWe take galangal sample after hot water bath treatment as material to extract flavone,and assess the influence of temperature of water bath,ethanol concentration,solid-liquid ratio and soak time on flavone extraction.And then,BBD response surface experiment was proceeded to optimize the flavone extraction,and temperature of water bath of 82?,ethanol concentration of 57%,soak time of 179 min,solid-liquid ratio of 1:40 were the optimized condition with yield of 99.39 mg/g and purity of 42.92%.Also,antioxidant experiment was carried out to compere of the antioxidant activity of flavone extracted from raw sample or residue,as the result,both of these samples show great antioxidant activity through DPPH·,ABTS·~+,superoxide anion free radical scavenging assay and Reduction capacity test,with little difference.(3)Flavone purification with macroporous resinEvaluating the capacity of static adsorption and desorption of 7 macroporous resins,we find that XDA-6 resin shows the better performance in static adsorption and desorption experiment than others,which was selected to purify galangal flavone.After that,we estimate static adsorption in difference temperature and desorption with difference concentration of ethanol as eluent,and find that the static adsorption of XDA-6 resin fit model of Langmuir,with no difference during 20~40?,and 70%(v:v)ethanol would be the proper eluent.Also,dynamic absorption and desorption were carried out and the best condition of purification were follow:flow velocity of 2 BV/h,sample concentration of 2mg/mL,sample volume of 31.6 BV,and the elution were proceeded with amount of 70%(v:v)ethanol as desorption solvent 3.1 BV,flow velocity of 2.5 mL/min.After purification of condition above,the purity of the sample increased from 43.55%to 85.42%with the better activity of DPPH·and superoxide anion free radicals scavenging and reducing ability,and show the same performance in ABTS·~+free radicals scavenging test,which mean purification could improve the antioxidant capacity.In a word,we gain a new method to extract galangal essential oil and flavone continuously,which could obtain us two functional components from half amount of galangal,and flavone shows better quality after purification.
