Single factor experiments and an orthogonal test were used to optimise the extraction conditions for polysaccharide from Kaempferia galanga L.(galangal).The composition in this crude product was determinated,and the content of protein was moved away by the method of TCA to get the galangal ploysaccarides(GPs),then deteced it composition.The structure of GPs was elucidated by UV spectrum and IR spectroscopy.The molecular weight distribution of GPs was determined by high performance gel-permeation chromatography,and the monosaccharide composition of this polysaccharides was determinated by ion chromatography.The final optimum extractions were as follows:extraction time of 6.5 h,water-to raw material ratio at 1:35,extraction temperature of 80?,extraction times of 1 times.Under this condition,the average yield of crude polysaccarides from ginger was 8.53%,and the crude polysaccharides contained 34.26%carbohydrates,16.40%protein.Following the protein was removed,the relatively pure product,named giner ploysaccarides(GPs),contained 75.20%carbohydrates,24.07%uronic acid.UV spectral analysis showed that GPs is substantially free of protein or nucleic acid.IR spectroscopy analysis found that GPs is an ?-type pyranose polysaccharide.The molecular weight of GPs was 8.5 x 105 Da,and the ploysaccarides was composed of fucose,rhamnose,arabinose,galactose,glucose,xylose,mannose,glucuronic acid and galacturonic acid.The molar ratio of them was 0.37:3.12:1.23:6.39:1.36:3.09:1.00:0.91:1.27 in turn.Building mice model of H22 to research the anti-tumor activity of GPs in vivo,and the result showed that GPs in tumor-bearing mice can effectively inhibit H22 cell proliferation,especially the results of the middle dose(200 mgkg-1d-1)and high dose(300 mgkg-1d-1)GPs group,displayed no significance different,played better than that of the low dose(100 mgkg-1d-1)group.In addition,the results of related assay including collection of the basic physiological indexes of the mice,detection of non-specific immune cell activities(intraperitoneal macrophage and splenic NK cell activity test),organ indicators analysis(organ index,HE staining,detection of lymphocyte subsets and spleen cell proliferation assay and NK killing assays)and blood test(blood routine examination and peripheral blood lymphocyte subsets determination),indicated that GPs play an antitumor effect through enhancing the autoimmunity of tumor-bearing mice,and the detection data of polysaccharide group was better than model group,and closer to the blank group.All in all,GPs is an ?-type pyran polysaccharide,and it plays a good anti-tumor effect on H22 cells in vivo.
