Blueberry is bilberry berry,which is rich in a large number of anthocyanin active substances,and has many physiological functions such as anti-oxidation,hypoglycemic,anti-inflammatory,anti-cancer,strong heart,softening blood vessels,anti-aging,and improving human immunity.In recent years,it has received a lot of attention.The efficient extraction of blueberry anthocyanins provides conditions for its powerful functional activity.This article focuses on the extraction,purification,structure identification,storage stability and in vitro antioxidative activity of anthocyanins from rabbit eyes of blueberry blueberry in Guizhou.It is provided for the further development and research of blueberry-related health products or drugs.Certain theoretical basis and basis.Through the study of this experiment,this paper obtains the following results:(1)Study on Extraction Technology of Blueberry Anthocyanin.The effects of the ratio of feed to liquid,volume fraction of ethanol,temperature,and extraction time on the yield of anthocyanin extraction were investigated.The response surface methodology was used to optimize the solvent extraction process to obtain the best The extraction protocol is a ratio of material to liquid 1:20(g/mL),pH=2 ethanol volume fraction 80%as extractant,temperature 30°C,time 30min,to extract twice better,Yield of 35.8.(2)Study on Purification Process of Blueberry Anthocyanidins.Experiments selected AB-8 macroporous resin fillers from four different types of macroporous resin fillers AB-8,LSA-12,LX-68,and D101 to purify blueberry anthocyanins.The anthocyanin static adsorption equilibrium time was 4.5 h,and the complete desorption time was 2 h.The macroporous resin was packed in a wet method.The column size was 2.4×30 cm,and the packed column height was 24 cm.The experimental results showed that the process conditions for the purification of blueberry anthocyanin with AB-8 macroporous resin were determined as follows:the anthocyanin concentration was At 3.5 mg/mL,the concentration of the desorption solution was 50%ethanol at pH=2,15 times the amount of resin eluted,2.5 times the elution time,and the flow rate was 1 mL/min.(3)Composition Analysis of Blueberry Anthocyanin.The experiments identified four major anthocyanins in rabbit eyes blueberry by high performance liquid chromatography coupled with high performance liquid chromatography-mass spectrometry.Four anthocyanin monomers were identified in rabbit eyes blueberry,respectively.-Glucoside,petunia-3-glucoside,cyanidin-3-arabinoside,delphin-3-arabinoside.(4)Study on Storage Stability of Blueberry Anthocyanidins.Through experimental research,it was found that the anthocyanin is stable under the condition of pH less than 3.0,has good thermal stability below 40°C,and has certain high temperature resistance in a short time.Sensitive to light,the anthocyanin is poor in resistance to oxidation and reduction,and ascorbic acid decreases the stability of the anthocyanin and accelerates the discoloration of the anthocyanin solution.Sodium sulfite has a good effect on the stability of blueberry anthocyanins.(5)Antioxidant Activity of Blueberry Anthocyanin.Using the anthocyanin purified material and ascorbic acid VC as antioxidant activity,the maximum clearance rate of blueberry anthocyanins to DPPH free radicals was up to 62.04%.The maximum clearance rate of blueberry anthocyanins to OH radicals was up to 85.12%,blueberry anthocyanin reduction capacity is lower than ascorbic acid V_C.
