Flavanones,also known as 2,3-dihydroflavones,is a chiral drug and one of the effective components of many medicinal plants.According to literature reports,many flavanones have bactericidal,anti-inflammatory,anti-tumor,anti-HIV and are a kind of medicines with high research value.China Food and Drug Administration clearly stipulates that the stereoisomers of chiral drugs should be obtained separately for necessary pharmacological studies.Therefore,chiral separation of flavanone enantiomers and acquisition of optical pure flavanone are of great significance.In this paper,flavanone enantiomers were separated by synchronous switched simulated moving bed(SMB)and its improved technology,asynchronous switched simulated moving bed.The main research contents and results are as follows:A method for the determination of enantiom ers of flavanones by high performance liquid chromatography was established;Column:Chiral column of Chiral OD,column size is 10×150 mm;Mobile phase:n-hexane/isopropanol=90/10(v/v);Column temperature:30 ?;Ultraviolet detection wavelength:254 nm.The mathematical model of Chiral OD column was established.The axial diffusion coefficient and total mass transfer coefficient were determined by empirical formula.Linear adsorption isotherms of flavanone enantiomers on Chiral OD column were determined using n-hexane/isopropanol(volume ratio 90/10)as mobile phase.Separation conditions of flavanone enantiomers in a three-zone simulated moving bed were designed using triangle theory.In the complete separation region of triangle theory,the operation conditions with feed flow rate of 0.25 mL/min were experimentally verified.The experimental results show that the purity of both the raffinate outlet and the extraction outlet reaches more than 99%.Linear decreasing inertial factor particle swarm optimization(LDIWPSO)was used to optimize the operating conditions of three-zone SMB with synchronous switching and three-zone SMB with asynchronous switching respectively.The experimental results show that the purity of the products under the two operation conditions is above 99%.In the optimization results,the zone configuration of synchronous switching SMB is 1/1/2,and the average zone configuration of asynchronous switching SMB is 1.1/1.35/1.55.The sample handling capacity of asynchronous switching SMB is 12.9%higher than that of synchronous switching SMB.
