Codonopsis pilosula is a party genus.Codonopsis pilosula polysaccharide has anti-tumor,anti-oxidation and immunity enhancement effects.At present,there are few systematic studies on the polysaccharides of Codonopsis pilosula.In this study,the water-ethanol system was used to extract Codonopsis polysaccharide from Codonopsis tangshen Oliv.,and the extraction technology was optimized by using response surface.In order to verify the versatility of the process,the amplification was verified and the extraction method was extended to the Codonopsis compound.The macroporous resins were used to purify the Codonopsis pilosula polysaccharide and the Codonopsis pilosula polysaccharide was further prepared into granules,thus obtaining a certain process parameter basis.The main contents of the research are as follows:The water-ethanol system was used to extract the polysaccharides from Codonopsis pilosula and Codonopsis compound.According to the single factor experiment,the response surface was used to optimize the extraction process.The results showed that the optimized extraction process: extraction temperature 64?,extraction time 108 min,ethanol concentration 46%,and the content of Codonopsis pilosula polysaccharide was(21.66 ±0.34)mg/g.The pilot scale-up process verified that the content of Codonopsis pilosula polysaccharide was 168.98 mg/g.The optimized extraction process of compound Codonopsis pilosula polysaccharides: extraction temperature 60?,extraction time 118 min,ethanol concentration 43%,compound Codonopsis polysaccharide content(26.32 ± 0.36)mg/g.The adsorption capacity of different resins was investigated.Then,the loading concentration,loading volume,aspect ratio and eluent concentration were determined.The separation and purification process was a loading concentration of 10 mg/mL,a loading volume of 160 mL,an aspect ratio of 1:8,and an eluent of 5% aqueous ethanol solution.The test was carried out with a pilot test to obtain a solid product of Codonopsis pilosula polysaccharides with a content of 25.12%.The study explored the cell experiment,reducing ability experiment and DPPH free radical scavenging experiment of Codonopsis polysaccharide and Codonopsis compound polysaccharide.The results showed that both Codonopsis pilosula polysaccharide and compound Codonopsis pilosula polysaccharide could inhibit the proliferation of cancer cells and have certain antioxidant activity in vitro.The mechanism showed that the compound Codonopsis pilosula polysaccharide induced apoptosis by activating the expression of cysteine protease.The study also explored the preparation process of Codonopsis pilosula polysaccharides.The response surface was used to optimize the dry and wet granulation process of Codonopsis pilosula polysaccharides.The results showed that the best method of dry granulation was as follows: the mixing ratio of ?-lactose and mannitol in the filler was 1:1.5,the ratio of extract powder to filler was 1:3,and the pressure was 8 MPa,and the comprehensive score was 96.00 ± 0.34.The best process for wet granulation was that the binder ethanol solution concentration was 62%,the ratio of extract powder to dextrin was 1:6.5,the drying temperature was 65?,and the comprehensive score was 91.60±0.28.In summary,this study obtained a complete set of Codonopsis pilosula polysaccharide extract extraction,purification and biological activity evaluation methods,and preliminary research on the process of Codonopsis pilosula polysaccharide instant granule preparation,which has important application value for the comprehensive development of Codonopsis pilosula polysaccharide.
