Camellia oleifera Abel seed cake,containing a lot of polysaccharides,and is a by-product during Camellia oleifera Abel seed oil processing.Therefore,the utilization of Camellia oleifera Abel seed cake polysaccharide has theoretical value and practical significance.In this study,crude polysaccharides extracted from Camellia oleifera Abel seed cake were used as raw material.The structure and antioxidant activities of the purified polysaccharide were determined.The main findings are as follows:?1?Camellia oleifera Abel seed cake was used as raw material to optimize the extraction process of polysaccharides.The best extraction process was ultrasonic time15 min;ratio of material to liquid 1:25;extraction temperature 70?;extraction time150 min.Under these conditions,the average extraction rate of polysaccharides reached16.76%.?2?Deproteinization process was optimized assisted with enzyme.Papain was chosen for the best enzyme employed in the deproteinization process.The best process was enzyme addition 3.1%,enzyme treatment temperature 57.5?,pH=6.0.Average removal rate of protein reached at 87.17%.?3?The crude polysaccharide was purified by DEAE-52 celllose anion exchange resin.Four polysaccharide components were obtained:SCP-a?water?,SCP-b?0.2 M?,SCP-s?0.4M?,SCP-c?0.8M?,respectively.SCP-s was further purified by Sephadex-G200,and the two components SCP-s1 and SCP-s2 were obtained.Ultraviolet spectral scanning shows that it contains almost no nucleic acids and proteins.Infrared spectroscopy?FT-IR?showed that they had characteristic absorption peaks of polysaccharides.The molecular weights of SCP-a,SCP-b,SCP-c SCP-s1 and SCP-s2 were 8.26×104 Da,2.05×107 Da,1.44×107 Da,2.00×104 Da and 7.16×106 Da,respectively.Mannose is a dominant monosaccharide with the largest proportion of monosaccharide composition in the five components of polysaccharides.The enthalpies of the five polysaccharides were 163.2,182.4.5,149.8,186.2 and 112.5 J/g,respectively.X-ray diffraction analysis showed that purified polysaccharides all exhibit semi-crystalline state.?4?The antioxidant activities of crude polysaccharide?CCP?and SCP-a,SCP-b,SCP-c were conducted in a dose-dependent manner.The four polysaccharides had significant scavenging capacity on ABTS and DPPH free radicals.The anti-lipid peroxidation ability and hydroxyl radical scavenging ability of polysaccharides showed synergistic consistency with that of the chelating ability of Fe2+.
