Supercritical fluid chromatography(SFC)is a separation technology similar to normal phase chromatography,using supercritical carbon dioxide fluid as mobile phase.It is an advanced analytical technology with the advantages of short analysis time,high sensitivity and environmentally friendly.It has been applied in many fields such as food,medicine,petrochemical industry and so on.In this paper,several chiral drugs were separated by supercritical fluid chromatography,and several saponins in Panax ginseng and Panax notoginseng were separated and determined.The main research contents are as follows:1.The chromatographic conditions of chiral separation of citalopram hydrobromide,trimebutine maleate,cetirizine hydrochloride and rosiglitazone maleate were determined by studying the factors of chromatographic column,modifier,additive,column temperature,back pressure and flow rate.The optimized chromatographic conditions were applied to the quality control of related preparations,controlling the chiral enantiomeric impurities.2.The chromatographic conditions for the simultaneous separation of ginsenoside Rg1,ginsenoside Re,ginsenoside Rb1 and Notoginsenoside R1 were determined through the study of chromatographic column,modifier,additive,column temperature,back pressure and flow rate.The four saponins in Panax ginseng and Panax notoginseng were separated and determined by this chromatographic condition,and the results were compared with those in the volume 1 of Pharmacopoeia of the People's Republic of China 2015,which shows no significant difference.Supercritical fluid chromatography provides a new simple,rapid and environmentally friendly method for chiral drug resolution,which can be used as a supplement to liquid chromatography or gas chromatography.Supercritical fluid chromatography uses non-toxic and low-cost carbon dioxide as the main mobile phase,which can save a lot of organic reagents,and has the characteristics of fast analysis and good separation effect.For traditional Chinese medicines with complex components,supercritical fluid chromatography can quickly separate many components of traditional Chinese medicine,which produces less waste liquid,is relatively environmentally friendly,saves costs,and achieves accurate results.
