In view of the problems that traumatic hemorrhage can not be quickly and effectively controlled,wound infection and wound healing is slow,etc.This study intends to develop a new type of medical dressing that can promote hemostasis,and has the efficacy of sustained antibacterial,and rapid wound healing.In this study,micro-nano fiber medical dressings blended with different mass ratios of silk fibroin(SF)/chitosan(CS)/halloysite nanotubes(HNTs)were successfully prepared by electrospinning.The morphology,structure and properties of the samples were studied by electron microscopy,universal material testing machine and thermogravimetric analyzer(TGA).The test results showed that the addition of HNTs significantly changed the microscopic morphology of the fibers,such as thicker diameters,and the fiber surface became rougher with the addition of HNTs.When the content of HNTs was less than 4%,HNTs could be dispersed in the nanofiber more uniformly.After changing the content of CS,the higher the content of CS was,the more concentrated the fiber distribution was,and the more it affected the fiber diameter.The study of mechanical properties showed that the addition of HNTs could improve the tensile properties of electrospun materials.When the mass ratio of SF to CS was 90:10,the tensile strength was increased from 0.44 ± 0.35 MPa to 2.23 ± 0.14 MPa,and the elastic modulus was increased from 0.16 ± 0.03 MPa to 0.64 ±0.06 MPa,which was expanded by about 5 times and 4 times compared with samples without HNTs.Respectively,and the elongation at break increased from 28.27 ± 7.22%to 44.74± 7.75%compared with samples without HNTs.In this study,the antibacter:ial drug chlorhexidine digluconate(CHD)was loaded into the hollow cavity or deposited on the outer wall of HNTs,and the drug-loaded SF/CS/HNTs blended electrospun materials were prepared.Results showed that the drug loading rate and encapsulation efficiency were 8.60%±1.11%and 61.47%± 8.19%respectively.The drug-loaded electrospinning materials exhibit excellent antibacterial properties against S.aureus and E.coli.In vitro whole blood coagulation time(WBCT)experiments showed that the addition of HNTs can quickly coagulate the blood of the wound,which is beneficial to the rapid wound healing.The drug release experiment showed that the release time of the drug model in our study could be prolonged by about 8 days when the drug-loaded HNTs were incorporated into electrospinning materials.In addition,the release rate of CHD in acidic environment was higher than that in neutral system,which is beneficial to wound healing in an acidic environment.In summary,electrospun micro-nanofiber medical dressings based on SF,CS,and HNTs have strong mechanical properties,hemostatic properties,and sustained antimicrobial properties.The drug loaded SF/CS/HNTs prepared in this study can be used as candidates for new medical dressings.
