Eucalyptus citriodora was one of the important forest tree species in GuangXi.In this paper,the extraction,purification and antioxidant activities of total flavonoids,polyphenols,polysaccharides and proteins from eucalyptus citriodora resin were studied.The main research contents and results were as follows:(1)Extraction,purification and antioxidant activities of flavonoids from eucalyptus citriodora resin.Firstly,the total:flavonoids in eucalyptus citriodora resin were qualitatively identified by hydrochloric acid-zinc powder and TLC,and then the quantitative analysis method for determination of flavonoids by UV spectrophotometry was established with rutin as the standard.The linear relationship between flavonoids concentration(C)and absorbance(A)was:A=25.11c+0.08.The method was used to determine the contents of crude flavonoids(A)in eucalyptus citriodora resin.Then chloroform-butyl acetate,chloroform-dimethylformamide,and dimethylformamide chloroform were used to extract the purified compounds B,C,and D.Their total flavonoids mass fractions were 10.39%,18.03%,and 44.15%.Finally,A,B,C,D has carried on antioxidant activities,The activities of A,B,C and D in scavenging 2,2-Diphenyl-1-picrylhydrazyl(DPPH·)were 91.75%,74.22%,88.44%,91.56%,total antioxidant capacity were 19.99,1.75,4.39,3.79 ?mol rutin equivalent(RE)·g-1,reducing power were 1.16,0.36,0.39,0.45 pmol RE·g-1.(2)Extraction,purification and antioxidant activities of polyphenols from eucalyptus citriodora resin.The polyphenols in eucalyptus citriodora resin were identified as hydrolyzed polyphenols by qualitative methods such as gelatin solution and ferric chloride solution.Using gallic acid as the standard substance,the quantitative analysis method of polyphenols was established by UV spectrophotometry,and the linear equation of concentration(C)and absorbance(A)was given as follows:A?145.30c+0.16.The contents of polyphenols in the resin was 13.40%.The mass fractions were 28.01%and 39.60%that eucalyptus citriodora resin polyphenols crude extract by ethyl acetate,purification,alcohol water sink-n-butyl alcohol purification.Purifications and vitamin C(Vc)were carried out antioxidant activity experiment and the results showed that they remove DPPH free radicals activities were 89.68%,86.45%,89.11%,91.12%,the total antioxidant activity were 13.44,12.68,15.46,14.89?mol,gallic acid equivalent(GE)·g-1,reducing power activity were 0.99,0.78,1.04,1.20?mol,GE·g-.(3)Extraction and purification of polysaccharides from eucalyptus citriodora resin and their antioxidant activities.The determination of total sugar by 3,5-dinitrosalicylic acid spectrophotometric method was established.The linear relationship between total sugar concentration(C)and absorbance(A)was as follows:A=4.38c-0.47.The total sugar contents of eucalyptus citriodora resin was 18.21%.The quantitative analysis method of polysaccharide was established by phenol-sulfuric acid spectrophotometry.The linear relationship between polysaccharide concentration(C)and absorbance(A)was:A=12.34c+0.05.The contents of polysaccharide in lemon eucalyptus was 16.78%.By Sevage method,trichloroacetic acid method and other methods for purification of polysaccharide of eucalyptus citriodora resin won the mass fraction of 22.14%,38.56%,51.66%respectively purified polysaccharide,and their antioxidant activities were studied,the results show that the antioxidant activities of different mass fraction of polysaccharide order:80%ethanol purification>60%ethanol purification>30%ethanol purification,the higher the mass fraction,the higher the antioxidant activity.(4)Extraction of protein from eucalyptus citriodora resin and their antioxidant activities.The quantitative analysis method of lipoprotein in eucalyptus citriodora resin was established with bovine serum protein as the standard,and the linear relationship between protein concentration(C)and absorbance(A)was given as:A=5.70c+0.02.The lipoprotein contents of eucalyptus citriodora resin was 7.47%.The results showed that the antioxidant capacity of the crude protein extract increased with the increase of concentration.
