Application Development Of Rapid Fluorescence Culture Detecting System BACTEC In Commercial Sterility Testing

Canned food commercial sterility refers to that the product does not contain pathogenic microorganisms or non-pathogenic microorganisms that can reproduce in the product under normal temperature after high temperature sterilization.Under this condition not only all pathogenic microorganism and the microorganisms that can form toxins have been damaged,but also other microorganisms that can grow in the product under room temperature storage conditions have been destroyed.As one of dairy products,due to its convenient preservation,long shelf life,UHT(ultra-high temperature)sterilized milk is favored by consumers.It adopts the commercial sterility detection standard.Based on the current GB4789.26-2013 standard and SN/T2100-2008 standard,this paper proposes an improved detection method for commercial sterility.This method customizes a detection bottle with no medium,inoculating different varieties and quantities of microorganisms in the bottle,Evaluate the specificity,sensitivity and applicability of rapid fluorescence culture detecting system BACTEC used in UHT milk commercial sterility.Build an improvement method used the Bactec system,compared with the traditional method of GB standard and SN standard.The verification results show that the rapid fluorescence culture detecting system can meet the requirements of rapid commercial sterility detection of UHT sterilized milk,which can shorten the detection time from 10 days to 3 days or less.It can accelerate the detection speed,reduce enterprise inventory.This method requires only one modified bottle,the process is simple,and the cost is saved.Moreover,it can clearly distinguish commercial sterility or sterility,and is highly consistent with the result of national standard(GB),and the result is more objective by reducing sensory judgment,and has a certain market application prospect.
