Licorice is a traditional Chinese medicine and an important raw material for modern medicine,food and cosmetics.The long-term industrial production only extracts the glycyrrhizin?also known as glycyrrhizic acid?in licorice as the main active ingredient,and the waste residue and residual liquid after extracting glycyrrhizic acid are abandoned,resulting in great environmental pollution and waste of resources.In this paper,the acid residue of licorice was used as the material.Firstly,the content of main chemical components in licorice residue was determined by national standard method.The alcohol-soluble small molecular substance in licorice residue was extracted by methanol,and high performance liquid chromatography was used.The chemical structure of the main chromatographic peaks in the extract was preliminarily identified by diode array detection-mass spectrometry?HPLC-DAD-MS?combined with standard methods,and its content was determined by HPLC.On this basis,the light licorice was used as the target,methanol was used as the extraction solvent,and the yield and purity of licorice were used as indicators.The traditional method of organic solvent extraction was used to extract the licorice from licorice residue.The effects of various factors on the extraction were investigated by single factor experiment.The optimal extraction conditions of the light licorice were determined by orthogonal experimental design.Then,the organic extract was used to extract the extract of Glycyrrhiza glabra L.,and the model organism C.elegans was selected to study the antioxidant activity of the main extract phase.At the same time,the hypolipidemic function of the main extract was carried out by using the hyperlipidemic rat model.Preliminary research.The dietary fiber powder of licorice residue with different particle sizes was treated by ultrafine pulverizer.The effects of water holding capacity,oil holding capacity and swelling of dietary fiber with different particle size licorice residues were determined.The influence of physical and chemical properties.The main findings are as follows:The content of dietary fiber of Glycyrrhiza glabra L.is about 70.15%;it is mainly insoluble dietary fiber,accounting for about 95%of total dietary fiber.In addition,it is also rich in starch and protein,with a content of 21.1%and 8.31%,respectively.Glycyrrhizin is an important alcohol-soluble component in Glycyrrhiza uralensis slag,the content is about 0.58%,and the purity in methanol extract is 8.96%.The optimal extraction process of licorice was as follows:extraction time 3 h,industrial methanol concentration 90%,extraction temperature 50?,ratio of material to liquid 1:35,extraction times 2 times,yield and purity were 0.477%and 8.94%,respectively.In the gradient extraction phase of the glycyrrhizin extract,the chloroform extraction phase yield was the highest,about 75%.Each concentration of chloroform extracts effectively prolonged the lifespan of C.elegans,increased the egg production of C.elegans,and prolonged the resistance of C.elegans under acute stress conditions.The 10?g/mL extract extended the lifespan of C.elegans by 43.84%,extending its lifespan under acute heat stress by 177%;100?g/mL extract prolonged the life of nematodes in an acute oxidative stress environment 60%,and increased the number of eggs laid by 52%.The chloroform extract of Glycyrrhiza uralensis extract can effectively treat hyperlipidemia in rats,reduce the levels of TC,CRE&UREA,AST&ALT in serum of hyperlipidemic model rats,increase HDL-C content,and maintain the dynamics of TG,ALB and TP.balance.The particle size has a significant effect on the water holding capacity,swelling property,oil absorption,ion exchange capacity and NO2-adsorption capacity of the licorice residue dietary fiber powder.The water-holding,swelling and ion-exchangeability are similar with the change of particle size,and the overall trend of particle size reduction is“first increase and then decrease”.When the particle size is75?m,the dietary fiber of licorice residue has the most expansion.Good,it is 0.27mL/g.When the particle size was 115?m,the licorice residue dietary fiber had the best water holding capacity and ion exchange capacity,which were 0.27 g and 1.1mmoL/g,respectively.The oil-holding property drops sharply after the particle size is less than 61?m,and the adsorption capacity of NO2-decreases with the decrease of the particle size.Based on the results of the above parameters,it is recommended that the dietary fiber of licorice residue be ultra-finely pulverized to a particle size of about 115?m and then used.
