Numerous of studies have demonstrated that soy protein digestion peptides have significant hypocholesterolemic activity.However,the mechanism of soybean protein in vivo hypocholesterolemic activity is not yet completely clear.This paper respectively used soy protein isolates and bean curd as raw materials,Alcalace protease and gastrointestinal simulation as enzyme solutions,macroporous resin,gel chromatography and centrifugal filter as separation means.And the products were analyzed its disrupt effect of micellar cholesterol transport for exploring the cholesterollowering activity of soybean protein hydrolysates.Both artificial cholesterol micelles and diluted natural swine bile were used as the micellar cholesterol delivering matrix,and both Kit and high performance liquid steamed-light scattering spectrometry(HPLC-ELSD)detection methods were employed to detect cholesterol content.The results indicated that the yield of Lactone Tofu Hydrolysate was(3 ± 0.1)%,which was far below the soybean protein isolate's(25 ± 2.0)%.The gastrointestinal simulation needed longer experimental cycle than Alcalace enzymolysis,but it was more familiar to body environment.The micelle cholesterol absorption rate of macroporous resin was(81.65±2.52)%.After separation of the gel chromatography,there were two components with the rate of(65.17±1.03)% and(76.24±1.45)% respectively.The MW(molecular weight)>10k of the Tofu protein hydrolysates had the strongest disruption effect for micellar cholesterol transportation.Its micelle cholesterol absorption rate was(62.40 ± 0.61)%,which was superior to(68.41±0.63)% of a specific sequence known peptide VAWWMY which was reported with high cholesterollowering activity.In a word,enzymatic hydrolysis of soybean peptides can disrupt micelle transporting cholesterol under the condition of this experiment.Thereby the cholesterol-lowering effect was indicated.
