The detection and intervention of intracellular temperature could provide the information about temperature and distribution of different regions and organelle in the cells,which can help us to understand the effect of temperature on the physiological activity of tumor and analyze the mechanism and effect of temperature-based tumor cell therapies.Therefore,it is significant to achieve sensitive,high-resoultion detection and regulation on tumor cells temperature.Aiming at this important requirement,Herein,a novel SERS nanothermometer which can be used as a sensitive nanothermometer with the function of active temperature control has been designed and prepared.The performance of the SERS nanometer thermometers was studied and their potential application in cells was preliminarily inverstigated.The specific chapters and contents are as follows:In the first charpter,the characteristics of tumor microenvironment and microenvironment temperature were briefly introduced,and then the importance of detecting the microenvironment temperature of tumor cells and the research progress of nano-thermometers were summarized.In addition,the advantages and applications of the gold nanoparticles with magnetic property,thermosensitive materials,and SERS technology were also discussed.Finally,the research contents and significance of this work were put forward.In the second chapter,the gold nanoparticles with magnetic property were synthesized and investigated.Firstly,the Fe3O4 nanoparticles were served as cores and sillica shells were synthesied on the cores via the hydrolysis of TEOS to obtain Fe3O4@SiO2 core-shell nanostructures.Then gold seeds were adsorbed on the surface of nanoparticles,fllowed by growing gold shells via seeds mediated growth,and the magnetic core gold shell nanoparticles?Fe3O4@SiO2@Au,GMNPs?with suitable size,good SERS enhancement effect and photothermal effect were obtained.In the third chapter,the SERS sensitive nanothermometers were prepared by in situ synthesizing thermosensitive polymer layer on the GMNPs and then assembling Raman molecules labelled gold nanoparticles onto the polymer coated GMNPs via electrostatic interaction.After systematically charecterizated the properties of the SERS nanothermometers,the preliminary intracellular application of SERS nanothermometer was carried out.
