Chiral sulfoxide is a kind of chemical intermediate that is widely used,and its synthesis has always been a hotspot in the field of chiral synthesis.At present,the production methods of chiral sulfoxides used in enterprises are mostly synthesized by applied chemical methods,but there are serious environmental pollution hazards in their production processes.In recent years,many scientists have focused on biocatalytic asymmetric synthesis of chiral sulfoxides.At present,there are few studies on the use of chiral sulfoxide thioether as a substrate.This study completed the biocatalytic preparation of omeprazole by the method of Soybean hull peroxidase catalysis,in line with the concept of green chemistry.In this paper,the PEG4000/citric acid buffer aqueous two-phase system was used to separate and purify the SHP in the bean shell.The influencing factors of the aqueous two-phase extraction were studied.The best lemon extracted by the two-phase system was investigated by single factor experiment.The effects of sodium concentration,PEG4000 concentration,aqueous phase pH and extraction time on the extraction efficiency were further optimized by using response surface design to optimize the optimal conditions for SHP extraction.Combined with acetone precipitation method,and further explored the mechanism of action of metal ion precipitation protein,the optimal Zn2+ion concentration for further purification of SHP is 0.01 mol/l,and the upper and lower limits of acetone fractionation are 2 volumes of acetone and one volume of acetone..Secondly,the subject studied the enzymatic properties of the laboratory-made Soybean hull peroxidase.The experiment found that the Soybean hull peroxidase is stable at a temperature of 20? to 50? and a pH of 4-8.Soybean peroxidase is not sensitive to metal cations and acid anions.Different organic solvents have different effects on their stability,and the stability in alcohol organic solvents is significantly higher than that of aliphatic hydrocarbons.Finally,this study used reverse micelle system to study the enzymatic reaction,and plotted the reverse micelle phase diagram to explore the factors affecting the asymmetric oxidation of SHP-catalyzed substrate unnatural omeprazole sulfide in reverse micelle system.The optimal organic solvent type,Wo value,CTAB concentration,enzyme amount,substrate concentration,reaction temperature and reaction time were determined to determine the optimal parameters of the reverse micelle system.Under the optimal conditions,the conversion rate of omeprazole thioether is 60%,and the ee value of s-omeprazole is over 96%.
