Reverse micelle extraction technology can not only get high quality oil and required functional properties of soybean protein in food industry,but also can separate soybean protein and oil simultaneously.Meanwhile,it can save resources,energy and overcome the shortcoming of the traditional separation technologies.Thus,it is of great significance to research the regulation of separation soybean protein and oil simultaneously by reverse micelle system.This paper tired to judge the dynamics of the extraction of soybean oil by AOT reverse micelle system through studying the regulation of oil extraction rate under main factors.And to get the properties of the oil extracted by AOT reverse micelle system,the research compared its physical and chemical properties with solvent extraction oil.The results showed that mass transfer process of extraction soybean oil by AOT reverse micelle was mainly in the first 20 minutes.The extraction process was in line with the internal diffusion control model,and its key step was the oil diffusion from the inside to the surface of particles.The apparent activation energy of extraction soybean oil by AOT reverse micelle system was 5.99 kJ/mol.The mass transfer model can be set up as 1+2(1-x)-3(1-x)2/3=0.248·exp(-720.8/T)·t.The oil extracted by reversed micelle system had smaller acid value,peroxide value and colour compared with the oil extracted by solvent.In order to study the regulation of the forward extraction of soybean protein by AOT reverse micelle system,the research extracted soybean protein under different factors in different levels,its structure,acting force and functional characteristics was investigated.The results showed that the extraction rate,the secondary structure,free-SH and disulfide bond content,emulsion stability and surface hydrophobicity of the soybean protein extracted tended to be stable when the extraction time was over 20 minutes.With short extraction time,0.04mol/L AOT,lower extraction temperature,the smaller molecular protein was dissolved in the reversed micelle system.And AOT concentration affected protein extraction and its functional characteristics mainly by changing the interface resistance.A certain amount of salt can promote the extraction of larger molecular protein,improved the stability of the protein structure and functional properties.The salt ions can not effectively promote the forward extraction process.When the pH was between 2 and 5,it dissolved mainly smaller molecular protein,when pH was between 6 and 8,there were more big molecular weight proteins while lack of medium molecular proteins.PH affected the structure and acting force mainly by changing the surface charge of soybean protein to affect the functional characteristics.Temperature mainly influenced the molecular inter-atomic forces to affect the structure and functional properties of protein.In the forward extraction,interfacial resistance,hydrophobic function and electrostatic force caused by pH was the key force to affect extraction.Based on the former researches,the regulation of the backward extraction of soybean protein by AOT reverse micelle system was researched.The results showed that:as the extension of extraction time,the extraction rate,and the secondary structure,free-SH and disulfide bond content,emulsion stability and surface hydrophobicity of the soybean protein extracted tended to be stable.The salt concentration did not have not much influence on the backward extraction but absolutely necessary.Salt affected the extraction rate,structure and function of protein mainly through influencing interfacial interaction.Interfacial resistance was the key force to influence backward extraction.The pH had a greater influence on the backward extraction,the soybean protein extracted had better function under weak alkaline environment and it was also the same when the temperature was und 40? and 50?.Free-SH content had a significant impact on the whole extraction process.
