Peony stamen is considered to be the essence of peony flower.It has beenfound to contain a variety of biologically active ingredients,which have antioxidant,anti-cancer,blood sugar lowering and blood fat lowering effects.With the growing awareness of dietary health,health foods containing natural active ingredients have become the new choice for modern people.Therefore,the peony stamen was used as the material to extract the peony stamen,and the anti-oxidation,hypoglycemic and hypolipidemic activities of the peony stamen extract were studied.The main results are as follows:1.The peony stamen was extracted by enzyme combined with ultrasonic method.The enzymatic amount,enzymatic pH,ultrasonic power and ultrasonic time were taken as the influencing factors,and the flavonoid extraction rate was the index.On the basis of single factor experiment,the extraction process was optimized by response surface method.The results showed that the maximum extraction rate was 0.95%under the conditions of 0.3%enzyme addition,pH=5,ultrasonic power 200W and ultrasonic treatment for 30 minute.The alcohol extract of peony stamen were purified by D101 resin,adsorbed by gas bath shaking,eluted with ethanol,concentrated and lyophilized for use.The results of high performance liquid chromatography showed that the main components of rutin,quercetin and paeoniflorin were 44.25%,15.5%and17%,respectively,and the total content was 76.75%.2.The antioxidant research on alcohol extract of peony stamen is divided into two parts:in vitro and cellular antioxidant.The in vitro antioxidant capacities were determined by reducing power and free radical scavenging capacity,respectively.The results showed that the mass concentration of alcohol extract of peony stamen was positively correlated with the scavenging ability of free radicals.The cell antioxidant model was established by H2O2-induced RAW264.7 cell and HUVEC cell injury.The experiment was divided into blank group,model group,control group and peony stamen dose group.Cell viability was measured by MTT assay,and the activity of various antioxidant enzymes in each group of cells and culture fluid was determined by biochemical colorimetry.The results showed that the cell damage was significant in the model group compared with the normal group,and the activities of antioxidant enzymes such as SOD,GPX,GSH and LDH in the cells were significantly increased,and the MDA content was significantly decreased.3.The mouse diabetes model was established by feeding the mice with high-fat and high-sugar diet to study the hypoglycemic and hypolipidemic ability of the ethanol extract of peony.The results showed that the diabetic mice showed symptoms of diabetes with“three more and one less”.Compared with the normal group,the glycemic index was significantly increased.After the administration of the peony extract,the glycemic index was significantly improved.The ethanol extract of peony stamen can increase the levels of TC,TG and HDL-C in serum and decrease the levels of ALT,AST,CREA and UREA.The concentration of TP and ALB in serum is also significantly increased.The organ index of the mice decreased significantly,and the tissue sections also showed that the ethanol extract of the peony stamen could inhibit the liver of the diabetic mice to a certain extent.
