The acceleration of industrialization has caused plenty of heavy metal ions to be discharged into the environment,causing serious pollution.These ions,such as mercury ions,are absorbed by the human body and will cause various diseases.However,some trace metal elements such as copper ions in the human body play a very important role in the regulation of physiological functions.Therefore,the detection of heavy metal ions is of great significance in the fields of environment,medicine and life safety.Based on the advantages of fluorescent probes,the fluorescence quenching fluorescence probe?CHT?for Cu2+ions and the ratiometric probe?PMH?for Hg2+ions were designed and synthesized by Schiff base reaction.The works are summarized in the following two parts:1.A fluorescent probe CHT for Cu2+ions was synthesized from coumarin derivative and2-benzothiazolinone oxime.Spectroscopic studies indicate that the probe has a specific fluorescence quenching response to Cu2+ions due to the formation of a 1:1 complex of the probe's heteroatoms with Cu2+.The sensitivity of the probe is excellent and the fluorescence intensity reaches minimized by adding 5 equivalents of Cu2+ions.The response time to copper ions can reach pleateau within 4 mins and maintain stable over a period of time.The probe has excellent selectivity and low toxicity to cells and can realize fluorescence imaging in cells.Finally,CHT can be used to detect Cu2+ions in the actual water sample.2.A ratiometric fluorescence probe PMH for identifying Hg2+ions was constructed by connecting phenanthroline and 3-methyl-2-benzothiazolinone.Spectroscopic studies show that the probe presents dual emission wavelength change in the addition of Hg2+ions.After the addition of Hg2+ions,the intensity of the emission peak at 445nm decreased,and the intensity at 520nm increased.The probe has a very fast response speed to Hg2+ions and has good stability.PMH has excellent selectivity for Hg2+ions and cannot be interfered by other ions.PMH can realize dual-channel fluorescence imaging of Hg2+ions in cells,and the probe successfully detect Hg2+ions in zebrafish,which has potential application in vivo detection.
