Actinidia arguta,a genus of Actinidia in Actinidiaceae,is a kind of small berries of lianas with cold resistance.The fruit of Actinidia arguta is sweet,sour and has unique aroma.The fruit of Actinidia arguta is rich in flavonoids,polysaccharides,triterpenes and other bioactive components.It has hypoglycemic,antioxidant,anti-tumor,anti-inflammatory and neurological effects.In this study,the fruit of Actinidia arguta was used as material to extract flavonoids and optimize the extraction process to determine the best extraction technology;On this basis,the inhibitory activity of flavonoids on α-glucosidase and antioxidant activity in vitro and the improvement of hyperglycemic level in hyperglycemic mice were studied.The main results are as follows:(1)Optimization of technological parameters for flavonoids extraction from Actinidia arguta.The flavonoids of Actinidia arguta fruit were extracted by ultrasonic assisted extraction,the main influencing factors were determined by single factor experiment.The response surface was optimized on the basis of single factor experiment.The optimum technological parameters of flavonoids in Actinidia arguta fruit were:ultrasonic power was 300 W,ultrasonic time was 6 min,ultrasonic temperature was 60℃,ethanol volume fraction was 60%,ratio of material to liquid was 1:25 g/mL.Under these conditions,the highest yield of flavonoids was 8.89±0.04 mg/g.(2)Study on the inhibitory activity and antioxidant activity of flavonoids from Actinidia arguta on α-glucosidase.The results showed that when the concentration of flavonoids was 1.0 mg/mL,the scavenging rate of flavonoids DPPH radical was 80.55%±2.21%,IC50=0.42 mg/mL;the scavenging rate of hydroxyl radical was 74.14%±2.78%,IC50=0.59 mg/mL.The inhibition rate of α-glucosidase was 73.28%± 1.69%,and IC50=1.87 mg/mL when the concentration of flavonoids was 10 mg/mL.The inhibitory type of flavonoids on α-glucosidase in Actinidia arguta was competitive,and the inhibitory effect was reversible.(3)Hypoglycemic activity of flavonoids from Actinidia arguta.Type Ⅱ diabetic mice model was induced by high fat and high sugar diet combined with streptozotocin(STZ),and the symptoms of diabetic mice were significantly improved by flavonoids of Actinidia arguta.The results showed that the flavonoids of Actinidia arguta could significantly reduce the fasting blood glucose level and serum insulin level in diabetic mice.To increase the expression level of glucokinase gene in the liver of diabetic mice so as to increase the activity of glucokinase in the liver of diabetic mice.In this paper,the functional activities of the active components in Actinidia arguta were studied in order to provide reference for the development and utilization of Actinidia arguta.
