Polyether ether ketone,as a new high performance thermoplastic polymer,is widely used in biomedical field,especially in orthopaedic implants,such as intercolumn fusion,hip joint repair and skull reconstruction,due to its excellent mechanical properties,chemical stability,good biocompatibility and radiopaque property.But due to its stable chemical properties of hydrophobic and low surface energy,make its have biological inertia,in the long run will produce fibrous tissue around the implant,cannot form integration with the surrounding tissue,prone to loosening and bone nonunion,so to improve its biological inertia,make its have biological activity,which has good integration and bone conduction performance,that has great significance in the field of PEEK biomedical application.Build porous structure in the surface of PEEK,will not only greatly increase the specific surface area,and provide suitable for cell growth space,three-dimensional space is favorable to the transport of nutrients and metabolites of discharge,allow the formation of blood vessels and bone ingrowth,eventually implant and bone tissue formation mechanical interlock,there is a huge contact area,and can prevent the implant loosening,with the surrounding bone tissue formation,thus improve the PEEK bone integration ability.In this project,the porous PEEK with hydroxyapatite active layer on the inner surface of the pore was constructed through the preparation of sodium chloride and hydroxyapatite composite pore-forming particles.The pore size distribution was 380?m?700?m,The three-dimensional space was not only suitable for cell growth,but also changes the chemical properties of the surface because of hydroxyapatite active layer on the inner surface.The water contact angle of PEEK loaded with hydroxyapatite(PEEK-HAp)is significantly lower than that of porous PEEK(PEEK-Porous),which indicates hydrophilia of PEEK has been improved,Through co-culture of mouse osteoblast precursor cells(MC3T3-E1),it has been found that PEEK-HAp has no cytotoxicity and has a good adhesion and proliferation performance relative to PEEK-Porous.The porous hollow hydroxyapatite on the inner surface of PEEK-HAp can be used as a drug sustained-release carrier to load the osteogenic drug icariin(ICA)and the antimicrobial drug moxifloxacin hydrochloride(MOX).Through in vitro drug release experiments,PEEK-HAp has certain sustained release effect on ICA and MOX,It also can be seen from the antibacterial experiment of the bacteria that the PEEK-HAp loaded MOX has good antibacterial performance.Through the expression of OCN,COLI and ALP of mouse osteoblast precursor cells,it is found that it has good osteogenic performance.Therefore,the design and application of PEEK porous structure can improve its biological inertia,make it have biological activity,good osteogenic and antibacterial properties,and hopefully solve the problem of its bone integration.Therefore,it has an active role in the application of biomedicine.
