In this paper,using Tremella fuciformis as raw material,the extraction and purification process of Tremella fuciformis polysaccharides by the composite enzyme-tannic acid method was determined by comparison.The antioxidant activity,hygroscopicity and moisturizing property of the extracted and purified Tremella Polysaccharides in vitro were studied.Finally,tremella polysaccharides were applied in moisturizing lipstick.The details are as follows:1.Extraction and purification of Tremella Polysaccharide by complex enzyme tannic acid methodFirst of all,three kinds of complex methods were designed to extract and purify tremella polysaccharide: quaternary ammonium salt complex method,salting out complex method and tannic acid complex method.The feasibility of this method was studied and the extraction process of compound enzyme tannic acid method of tremella polysaccharide was studied.It was proved that the extraction and purification of Tremella Polysaccharide by complex enzyme tannic acid method was feasible.Using single factor and orthogonal experimental design,it is proved that the optimal process of tannin extraction and purification of Tremella Polysaccharide by complex enzyme tannic acid method is: the concentration of tannic acid is 40 g / L,the reaction temperature is 80 ?,the reaction time is 50 min,and the primary and secondary factors affecting the extraction rate of polysaccharide are reaction time >tannic acid concentration > reaction temperature.The yield is 21.76%,protein removal rate is 31.12%,polysaccharide loss rate is 15.06%,which is 0.53 times of the time of beating + enzyme isoelectric point method.2.Study on the activity of Tremella fuciformis polysaccharidesTo explore the anti-oxidation ability of tremella polysaccharide,the scavenging ability of tremella polysaccharide to DPPH free radical and superoxide anion free radical was tested.In the experiment of eliminating the former,when the solution concentration is 8mg / ml,the clearance rate of Tremella Polysaccharide sample is55.25%;in the test of eliminating superoxide anion radical,when the solution concentration is 8mg / ml,the clearance rate is 46.53%.Conclusion: tremella polysaccharide has the ability of antioxidation.The moisture absorption and moisturizing properties of Tremella fuciformis polysaccharides in vitro were determined.The hygroscopicity of Tremella fuciformis polysaccharides was tested in the humidity environment of RH=43% and RH=81%,and trehalose and glycerol were used as control.From the experimental results,it can be seen that the hygroscopicity of Tremella fuciformis polysaccharides was better than trehalose,slightly worse than glycerol,and the comparison of the moisturizing performance was: glycerol > Tremella Polysaccharides > trehalose.In the environment of dry silica gel and RH=30%,the moisturizing properties were compared: glycerol >Tremella Polysaccharides > trehalose.3.Formulation design and quality evaluation of Tremella fuciformis moisturizing polysaccharide lipstickAccording to the characteristics of lipstick and the design principles of moisturizing lipstick products,based on the basic formula of lipstick,the influence of wax composition and temperature on the quality of lipstick was discussed,and the best wax composition was determined.The proportion of white beeswax,candlelight tree wax and palm wax is 10:1:1.The results of orthogonal test were used to study the effect of the ratio of oil wax and grease on lipstick quality.The best ratio was 1:3:1 of mixed wax,olive oil and lanolin.The texture of lipstick made with this ratio is suitable in soft hardness and good in moisture.After the optimal formula is determined,the moisture retention of different concentrations of tremella polysaccharide is tested.The experiment shows that when the concentration of tremella polysaccharide is 1%,the moisture retention of lipstick is the best.
