Maize,wheat,barley and other food crops in the process of growth or storage will be contaminated with mycotoxin.Toxins not only affect the quality of food,but also harm human health.Fumonisins and corn gibberelone both belong to a kind of mycotoxin.The reference material has a high degree of uniformity,good stability and accurate characteristic value and can reproduce and transfer,so as to ensure the continuity and comparability of the measured results in time and space,and thus ensure the accuracy of the measured results.Therefore,the development of reference materials has been widely recognized at home and abroad.There is a big gap between China and developed countries in the development level of fungus-toxin matrix reference materials,so we have to rely on imports to meet the testing needs of domestic laboratories.This takes a long terms and wastes a lot of money,which is not conducive to the development of China's testing level.Based on this,the development of two matrix reference?quality control?materials of Fumonisin FB1 and Zearalenone ZEA in corn flour was carried out.The main research contents are as follows:?1?Commercially available corn particles were used as raw materials.After screening,add the standard solution and soak,freeze-drying,milling,blending and vacuum sealing,the uniformity and stability of standard materials were investigated,and the contents were determined in cooperation with several laboratories.Finally,the uncertainty of samples was evaluated.?2?Ultra-high performance liquid chromatography-tandem mass spectrometry?UPLC-MS/MS?was used to test the homogeneity of the samples,and the data were statistically analyzed by one-way analysis of variance.The results showed that the F values of the two reference materials were less than the value of F0.05,that is,the uniformity of the two samples met the requirements of the reference materials.?3?The short-term stability and the long-term stability of the two reference materials were investigated for 14 days and 6 months,respectively.The test method used is the same as the uniformity test method.The measured data are fitted with a linear model and the slope is used to evaluate the stability of the material.The results show that there is no significant change in the characteristic value of the two reference substances in the monitoring time,it has good stability in the short-term and long-term investigation period.?4?The authoritative and qualified laboratory was selected for joint determination to ensure that the test methods used by all laboratories are consistent.After statistical analysis of the detection results and elimination of suspicious values,the average value of the retained data is the characteristic value of the reference materials.The characteristic value of matrix reference materials of Fumonisin FB1 in corn flour was 1476?g kg-1,and that of Zearalenone ZEA in corn flour was 137?g kg-1.?5?The uncertainty caused by the process of inhomogeneity,instability and determination is evaluated and the total uncertainty is obtained.The characteristic value and uncertainty of matrix reference materials of Fumonisin FB1 in corn flour were 146?237?g kg-1 and that of Zearalenone ZEA in corn flour were 137?26?g kg-1.In summary,the two matrix reference materials of Fumonisin FB1 and Zearalenone ZEA can be used as quality control samples.Then can provide technical support for instrument calibration,operator level assessment and laboratory quality control in the process of toxin detection in China.
