Chicken meat is a common source of animal food on the daily table.It not only has the advantages of high protein,low fat and low cholesterol,but also tender meat,delicious taste and rich nutrition.With the continuous expansion of the poultry farming industry and the increase in farming density and breeding efficiency,the abuse and residue of veterinary drugs have become a hot topic for consumers.In order to ensure the quality of chicken products on the market,it is necessary to strengthen the effective detection of veterinary drug residues in chicken.Surface Enhanced Raman Spectroscopy(SERS),as a fast,simple,and highly sensitive detection method,has its unique advantages in the field of animal residue detection.In this paper,three kinds of antibiotics,norfloxacin,enrofloxacin and ciprofloxacin,were detected by surface enhanced Raman spectroscopy(SERS)in order to explore the feasibility of rapid detection of antibiotic content(norfloxacin,enrofloxacin,ciprofloxacin)in chicken meat by surface enhanced Raman spectroscopy(SERS),study the spectral changes of antibiotics in chicken meat,and develop the detection of antibiotic residues in chicken meat In order to provide an important reference for the detection of antibiotics in chicken meat.The main research contents were as follows:(1)Rapid detection of norfloxacin content in chicken meat based on SERS technologyThe SERS technology was used to analyze the norfloxacin content in the chicken meat extract spiked samples,and the concentration of norfloxacin content in the chicken meat was obtained according to the Raman characteristic peaks at 728,1312 and 1442 cm-1.Then,the pre-processed spectra were respectively established based on the full spectrum of norfloxacin in chicken meat prediction model and based on the characteristic peak prediction model,and realized the prediction of norfloxacin content in chicken meat based on SERS technology.The experimental results show that:? The lowest detection concentration of norfloxacin in chicken meat could be detected by this method up to 0.1 mg/L.?The results of PLSR established after SNV pretreatment based on the whole band were the best.The results were Rc=0.9724,Rp=0.9371,RMSEC=0.0615,RMSEP=0.0959.? The S-G-CARS and SNV-VCPA methods based on the characteristic band could be used to predict the content of norfloxacin in chicken meat.The results were Rc=0.7883,RP=0.8991,RMSEC=3.1726,RMSEP=8.7063,RC=0.9988,RP=0.9977,RMSEC=0.0027,RMSEP=0.0152.(2)Rapid detection of enrofloxacin content in chicken meat based on SERS technologyThe SERS technique was used to analyze the content of enrofloxacin in the chicken meat extract,and the concentration of norfloxacin in the chicken meat was obtained according to the peak intensity of Raman characteristic peaks at 732,1320 and 1437 cm-1.After that,the pre-processed spectra were respectively established based on the full spectrum of enrofloxacin in chicken meat prediction model and based on the characteristic peak prediction model,and realized the prediction of enrofloxacin content in chicken meat based on SERS technology.The experimental results show that:?The lowest concentration of enrofloxacin in chicken meat could be detected by this method up to 0.05 mg/L.?The results of PLSR established after De-trending pretreatment based on the whole band were the best.The results were Rc=0.9982,Rp=0.9974,RMSEC=2.6811,RMSEP=6.1395.? De-trending-CARS and S-G-VCPA methods based on characteristic bands could be used to predict the content of enrofloxacin in chicken meat.The results were Rc=0.9999,RP=0.9999,RMSEC=RMSEC=0.0219,RMSEP=0.6695,Rc=0.9999,Rp=0.9999,RMSEC=0.0095,RMSEP=0.0185.(3)Research on the rapid detection of ciprofloxacin content in chicken meat based on SERS technologyThe SERS technique was used to analyze the content of ciprofloxacin in the chicken meat extract,and the concentration of norfloxacin in the chicken meat was obtained according to the Raman characteristic peaks of 714,1320,1394 and 1439 cm-1.The experimental results show that:?The minimum detection concentration of ciprofloxacin in chicken meat by this method could reach 0.2 mg/L.XThe results of PLSR established after De-trending pretreatment based on the whole band were the best,and the results were Rc=0.9979,Rp=0.9934,RMSEC=2.3126,RMSEP=6.235.? Based on the characteristic band,the SNV-CARS and S-G-VCPA methods could be used to predict the content of ciprofloxacin in chicken meat.The results were Rc=0.9986,RP=0.9997,RMSEC=0.0094,RMSEP=0.0404,Rc=0.9999,RP=0.9999,RMSEC=0.0032,RMSEP=0.0079.The results of the study show that it is feasible to detect norfloxacin,enrofloxacin and ciprofloxacin in chicken meat by using SERS technology.
