Walnut(Juglans regia L.)green peel,also known as green dragon clothing,is the outer green peel part of walnut,and is the main by-product of walnut initial processing.According to previous research reports,walnut peel peel contains walnut bioquinone,?-sitosterol,quercetin,vanillic acid,kaempferol,and walnuts and other biologically active chemical substances,and it has the functions of detoxification,heat clearing,antibacterial and analgesic.In addition,the walnut green peel peel can also inhibit the growth of E.coli,Staphylococcus aureus,and Bacillus subtilis in vitro;its volatile oil component has asthma and antitumor effects.The walnut green peel has many functional effects,but the lack of convenient and effective extraction technology,the walnut green peel peel is extremely wasteful every year and pollutes the environment.Therefore,this study intends to use walnut green peel as raw material,adopt ultrasonic-assisted extraction method,use different solvents such as water,ethanol,ethyl acetate,n-hexane to extract the chemical components of walnut green peel,respectively,and analyze by high performance liquid chromatography mass spectrometry.The effects of different solvents on the chemical composition of the extract were compared and analyzed;the experimental diarrhea model of the mouse was established by using the senna decoction solution,and the antidiarrheal effect of walnut green peel extracts with different solvents was compared and observed simultaneously.The diarrhea index,capillary permeability,expressions of cytokines IL-6,IL-12,TNF-?,and hydrophobic related genes AQP3 and AQP4 mRNA were recorded.The purpose of this study was to investigate the differences in the composition of walnut peel extracts obtained by using different solvents as extractants,and the antidiarrheal effect of walnut peel extracts on acute diarrhea model test mice,in order to provide experimental data support for the development of natural antidiarrheal products.The main findings are as follows:1.Preparation of different solvent extracts of walnut green peel and their chemical composition.The walnut green peel peel powder obtained after being dried and crushed and sieved through a 40-mesh sieve was used as the raw material,and extracted by ultrasonic-assisted extraction with different solvents.The supernatant was concentrated by rotary evaporation and freeze-dried under vacuum to obtain different walnut green peel extracts.The chemical constituents in walnut peel extracts were analyzed by HPLC-MS.The results showed that in the extract of walnut green peel,neochlorogenic acid,epigallocatechin gallate,isochlorogenic acid B,isochlorogenic acid A,quercetin,and chlorophyll were identified by HPLC-MS / MS.13 compounds,such as rosmarinic acid,kaempferin,isoquercetin,chlorophyllin,baicalein,phlorizin,walnut quinone,cinnamic acid,walnut green peel n-hexane extract and walnut green peel ethyl acetate extract Has the highest extraction rate,respectively 224.08 mg /g,224.18 mg / g;walnut green peel water extract is 113.39 mg / g,walnut green peel ethanol extract is 158.00 mg / g;main effective functions in walnut green peel The content of walnut quinone is the highest in n-hexane extract of walnut green peel,which can reach 103.53 mg / g.It shows that the order of the solvent effect of the active ingredient extraction of walnut green peel is: n-hexane> ethyl acetate> ethanol> water.2.Antidiarrheal effect of different solvent extracts of wild pecan green peel on miceKunming mice were administrated with senna extract solution to induce acute diarrhea.Then the walnut green peel peel water extract,ethanol extract,ethyl acetate extract and n-hexane extract were intervened at 100 mg / kg.bw to diarrhea mice.The experimental results show that hexane extract of walnut green peel has better diarrhea index reduction effect than water extract,ethyl acetate extract and ethanol extract of walnut green peel;walnut pecan n-hexane extract can also better reduce Peritoneal capillary permeability in mice,and the results of serum cytokine tests also showed that walnut peel peel n-hexane extract can better reduce the levels of inflammatory cytokines IL-6,IL-12,TNF-?;further qPCR The results also showed that diarrhea would lead to a decrease in the mRNA expression of jejunum AQP3 and ileum AQP4 in mice,and walnut peel peel n-hexane extract could more strongly inhibit the decline of these two expressions.It can be seen that walnut peel extract has a good antidiarrheal effect,and n-hexane extract has better effects than water extract,ethyl acetate extract and ethanol extract,and n-hexane extract has better effect.The reason may be that more active substances with antidiarrheal effect can be extracted from n-hexane.Therefore,walnut green peel is a natural potential raw material with antidiarrheal effect.
