Object In order to prevent the formation of abdominal adhesion more effectively and improve the physical properties of the surface coated mesh,Sirolimus was grafted onto the surface of polypropylene mesh by chemical reaction to prepare a novel sirolimus drug-coated polypropylene?SRL-PP?mesh.The physical properties of SRL-PP mesh was studied and its anti-adhesion property was studied by animal experiments.Methods The polypropylene mesh is treated with a plasma cleaner to form new functional groups on the surface,which significantly increases the surface activity of the material,and then graft the polyethylene glycol onto the surface of the polypropylene mesh,finally,sirolimus was grafted onto the surface of the mesh by chemical reaction to prepare the SRL-PP mesh,compared with polypropylene?PP?mesh and polyethylene glycol modified polypropylene?PEG-PP?mesh.The three groups of meshes were characterized and analyzed respectively?infrared spectrum detection,contact angle measurement?,and their tensile properties were tested.The three groups of meshes were implanted into the abdominal cavity of rats.The anti-adhesion properties of the new sirolimus drug-coated mesh were studied by observing the intra-abdominal adhesion and histopathological changes?hematoxylin-eosin staining and immunohistochemical staining?in rats.Results Infrared spectrum analysis showed that SRL-PP mesh had a new absorption peak at the wavelength of 1 643 cm-1?amide group?,suggesting that sirolimus was successfully uploaded to the surface of the mesh;SRL-PP mesh had excellent hydrophilicity,which provided favorable conditions for peritoneal mesothelial cell growth;the mechanical properties of SRL-PP mesh were not significantly different from those of PP mesh and PEG-PP mesh,which provided a good mechanical guarantee for clinical application.The degree of abdominal adhesion in SRL-PP group[1.00±0.58]was significantly lower than that in PP group[5.17±0.69]and PEG-PP group[4.00±0.58],and there was a significant difference between SRL-PP group and PP group?t=2.76,P<0.05?.The number of inflammatory cells and the expression of inflammatory cytokines in SRL-PP group were significantly lower than those in PP group and PEG-PP group.Conclusion The sirolimus was successfully loaded onto the surface of the polypropylene mesh by chemical reaction.Animal experiments showed that the sirolimus drug-coated mesh can significantly alleviate the abdominal adhesion in rats,which provides a basis for clinical trials and applications.
