Diabetic kidney disease(DKD)is a chronic kidney disease,which is mainly transformed from diabetes.It is also the most important and severe microvascular complication of diabetes.Borniotis chalcogenes originated in the novels of the former Soviet Nobel Prize winners in literature,and has since attracted attention.At the end of the twentieth century,Porphyromonas birchii entered the research focus of China.In 1993,Dr.Jia Chunbao,a well-known Chinese medicine practitioner,faded into research and made a major breakthrough.It was confirmed that Porphyromonas velutipes is a kind of fungi that can be used as medicine and food.Polysaccharides are one of the important ingredients for its pharmacological activity.In this paper,a high glucose,high fat diet and STZ(Streptozocin,STZ)combined induction method was used to construct a mouse model of diabetic kidney disease.The basic growth status,blood glucose,Studies on the effects of blood lipids,renal function,anti-oxidation indicators,and renal pathological observations,etc.The preliminary investigation of the mechanism of action of P.chrysogenum polysaccharides in the prevention and treatment of DKD from kidney inflammatory factors.The main results are as follows:(1)Effect of Polysaccharides from Porphyromonas obliquus on the growth indexes of DKD miceThe results show that IOP can significantly reduce the body weight and kidney index of DKD mice(P<0.01),and restore the growth of mice.Compared with the model group,the high-dose(480mg/kg)polysaccharide from Porphyromonas campestris had the best effect,reducing the weight of mice by 17.4%,the kidney index by 15.8%,and the mental state of mice tended to be normal.It shows that IOP can significantly improve the body weight,kidney enlargement and poor living conditions of DKD mice.(2)Effect of Polysaccharides from Porphyromonas obliquus on Biochemical Indexes of DKD MiceStudies have found that Porphyromonas betulina polysaccharide can significantly increase the symptoms of hyperglycemia,hyperlipidemia,renal function and renal structural damage in DKD mice(P<0.01).Compared with the model group,the high-dose(480mg/kg)Polysaccharides from Porphyromonas velutipes reduced TC levels in mice by 32.3%,TG by 53.8%,LDL-C by 42.5%,and HDL-C by 24%,Reduced the blood glucose,Scr,BUN and urine protein levels of mice,the glomerular structure disorder and ambiguity were suppressed.The results indicated that Porphyromonas velutipes polysaccharide has the ability to alleviate abnormal lipid metabolism disorders and repair renal function damage in DKD mice.(3)Effect of Porphyromonas polysaccharide on antioxidant indexes of DKD miceIt was found that the polysaccharide from Porphyromonas velutipes could significantly improve the weakened antioxidant capacity of DKD mice(P<0.01).Compared with the model group,the high-dose(480mg/kg)polysaccharides of Porphyromonas velutii increased the T-AOC content of mice by 56.4%%,T-SOD by 29.8%,GSH-Px by 27.9%,CAT content increased by 23.8% and MDA content decreased by 14.9%.These results indicate that the polysaccharide from Porphyromonas velutipes can enhance the ability of mice to resist lipid peroxidation.(4)Effect of Porphyromonas polysaccharide on inflammatory cytokines in kidney of DKD miceThe results show that high-dose(480mg/kg)polysaccharide from Porphyromonas betulinus can significantly inhibit the content of renal inflammatory factors.Compared with the model group,the levels of TNF-? and IL-6 decreased by 33.1% and 43.5%,respectively(P<0.01).The results indicated that Porphyromonas polysaccharide can relieve kidney inflammation and repair kidney damage in DKD mice by regulating the content of kidney inflammatory factors.To sum up,it is concluded that the polysaccharide from Porphyromonas velutipes has protective effect on kidney injury in DKD mice.
