The mulberry nutrients and active ingredients vary depending on the growing region and variety.The mulberry extract has been reported to have anti-oxidation,hypoglycemic,anti-cancer effects and is an excellent raw material for the development of functional foods.In this study,based on the analysis of nutrients and active components of three mulberry varieties,the mulberry polyphenol purified with macroporous resin was optimized to prepare mulberry polyphenol powder.The inflammatory cell model of mouse macrophage RAW264.7 induced by lipopolysaccharide was established to investigate the inhibitory effect of mulberry polyphenol on inflammatory cytokines and its anti-inflammatory mechanism.The main findings are as follows:1.Analysis of nutrient and active components of three varieties of mulberry.The contents of ash,water,soluble solids,crude protein,amino acid,polyphenols,flavones,polysaccharides and anthocyanins of Xinjiang Black mulberry(XJBM),Xinjiang White mulberry(XJWM)and Yueshen Da10(YSD10)were compared.The results showed that the contents of ash,crude protein,total soluble solids and total polysaccharide of XJBM and XJWM were superior to those from YSD10.However,the content of total polyphenols,flavonoids and anthocyanins of YSD10 had advantages.The in vitro antioxidant capacity test of 3 mulberry varieties showed that the antioxidant capacity of XJBM and YSD10 was superior to XJWM.2.Optimization of mulberry polyphenol refining process.Taking the purification effect of mulberry polyphenol as an index,the X-5 resin with the best adsorption capacity and desorption effect was selected.The optimal process for the purification of mulberry polyphenol was as follows: the crude extract of mulberry polyphenol with the concentration of 1.8mg/m L was loaded with 10 BV at 1.0m L/min.After adsorption equilibrium,the samples were eluted with a small amount of distilled water,and then eluted with 70% ethanol at 1.5m L/min to 4 BV.After refining,the polyphenol content in the sample increased from 1.9% to 41.2%.The purification effect was obvious.3.Anti-inflammatory activity evaluation of Mulberry polyphenol and its mechanism.The results indicated that the polyphenols of XJBM could inhibit the release of NO and the production of pro-inflammatory cytokines IL-1,IL-6 and TNF-? at 0-50 ?g/m L.But the anti-inflammatory activity of the polyphenols of YSD10 was not obvious.Polyphenols of XJBM may play an anti-inflammatory effect by inhibiting the phosphorylation level of p38 protein and then inhibiting the inflammatory signal transduction in MAPK signaling pathway.It may also have antiinflammatory effects by inhibiting I?B? degradation and p65 protein phosphorylation,preventing NF-?B dimer nuclear transport,and thereby inhibiting downstream gene expression of NF-?B signaling pathway.
