With the rapid development of social economy and the increase of people's pressure,there is more and more demand for alcohol consumption,and the physical harm caused by alcoholism is more and more serious.Tea saponin is a pentacyclic triterpenoid substance,which can stimulate the activity of alcohol metabolizing enzyme ADH and ALDH,inhibit inflammatory damage,relieve alcohol,and protect stomach and liver.However,there are still doubts about the safety of tea saponin,and the effect of antioxidant damage is not good.If the single product is made,its clinical application is limited.Therefore,the purpose of this study is to make up for the deficiency of tea saponin in the aspect of alcohol and liver protection,to find out the compound substance with tea saponin in the medicine and food homologous alcohol resolving substance,and to explore the mechanism of alcohol and liver protection,and to verify its safety to provide theoretical basis for the research and development of tea saponin alcohol resolving products.The main research contents are as follows:?1?Among the eight medicine and food homologous antidotes.The optimal ratio of tea saponin to Hovenia dulcis was 1:1,which was determined by the number of drunk,the number of dead,the time of tailing,the time of tolerance and the time of sobering up.The model of acute alcoholism in mice was established by intragastric administration of 0.16 ml/10g 52 degree Hongxing Erguotou.It can activate ADH and ALDH.It is suggested that the mechanism of tea saponin complex is to alleviate acute alcoholism by activating ADH and ALDH.?2?The model of chronic alcoholism was established by intragastric administration of 0.12 ml/10g 52 degree Hongxing Erguotou to mice every day.The results showed that the weight of mice after the intervention of tea saponin complex increased,the liver index decreased,and the high dose group showed significant difference.Combined with the HE staining pathology figure,the edema and blood filling phenomenon decreased,the degree of liver swelling relieved,and the morphology of liver cells showed integrity.Serum biochemical indexes alt,AST,TBIL and DBIL all decreased in different degrees,and the intervention effect was better.In the liver,especially in the high dose group,SOD and GSH-Px contents increased significantly,MDA contents decreased significantly.The results showed that the compound of tea saponin could effectively alleviate the harm of chronic alcoholism,and the antioxidant damage was good.?3?The safety test of the tea saponin compound was carried out.In the acute toxicity test,the LD50of acute toxicity test of mice is more than 5 g/kg·BW which belongs to the actual no poison.In the subchronic test,normal indicators were observed in mice with bright fur,active alertness and normal diet and excretion.There was no significant difference in body weight and food utilization between the groups and the blank group?P<0.05?.In the routine blood examination,the number of red blood cells,percentage of mononuclear cells and percentage of neutrophils in the high-dose group showed significant differences compared with the blank group?p<0.05?,while the other groups showed no significant changes.In routine urine test,there was no significant difference between the dose group and the blank group?p>0.05?.Compared with the blank group,the lung in the high dose group showed significant differences in organ weight and organ coefficient?P<0.05?.HE staining showed that there was a small amount of neutrophil infiltration in the alveolar wall of the small and medium-sized rats in the high dose group,which indicated that the dose of 500 mg/kg·BW was harmful to the mice,and the other doses were relatively safe.
