| Nature stellerite is a kind of low temperature hydrothermal type mine which content is abundent in Guangxi.And the extraction process is simple.In the laboratory,the stellerite crude ore is often made into zeolite molecular sieve by hydrothermal reaction.Zeolite molecular sieve is a kind of silicate or aluminosilicate crystal material which possess strong ion exchange ability.And composed by regular microporous and tetrahedral skeleton elements.The zeolite molecular sieves are interconnected by oxygen atoms to form a three-dimensional regular pore structure.Moreover,the size,shape selectivity and ion exchange of the molecular sieve were determined by the pore property.The OD value is an important evaluation index usually used evaluate the status of bacterial.In this paper,the growth curve of bacteria effected by P-type stellerite molecular sieves antibacterial was measured by microporosity plate.The antibacterial effect was studied.The experimental results show that the micropore gradient dilution method is less suitable and the detection speed is faster than that of the tube diluti on method.The tube dilution method indicated that the minimum inhibitory concentrations of antimicrobial agents against Escherichia coli,Staphylococcus aureus and Bacillus subtilis were between 24 mg/mL and 48 mg/mL,and the minimum bactericidal concentrations were 48 mg/mL to 96 mg/mL.However,the micropore gradient dilution method manifest that the minimum inhibitory concentration of Escherichia coli and Bacillus subtilis was between 24 mg/mL and 48mg/mL,and the minimum inhibitory concentration of Staphylococcus aureus was between 12 and 24 mg/mL.In addition,The minimum bactericidal concentrations of the three strains were between 48 mg/mL and 96 mg/mL.At the same time,it can concluded that the inhibitory effect of P-typestellerite molecular sieves antibacterial on Staphylococcus aureus was the best,and the inhibitory rate was 89%.The cultured E.coli,Staphylococcus aureus and Bacillus subtilis were treated by ultrasonic and freeze-thawing to break down the bacterial cells,and the cell sap was extracted for specific detection,and the cell sap was measured to contain the target protein.The effects of potassium formate to the target protein were investigated.The p eak surface figureof the protein was obtained by high performance liquid chromatogra phy(HPLC).The antibacterial mechanism was vertification by comparing the structure and content of the enzyme variation in intracellular.Indicated that the P-type zeolite molecular sieves Release of potassium formate as the main antimicrobial agent,and potassium dicarboxylate affects and changes the structure and traits of bacterial proteins after passage through the cell membrane,reduces the activity of bacteria,thus affects the work of bacterial proteins and makes it impossible to grow normally. |
