As a type of malignant tumor,Leukemia imposes a huge burden on patients and society.Therefore,it has considerable research value for the diagnosis of leukemia and minimal residual disease monitoring.The current research results reveal that the fusion gene BCR-ABL and its corresponding fusion protein are directly related to chronic myeloid leukemia,and can be used as one of the biomarkers for diagnosis.More and more attention has been paid to the SERS(Surface enhanced Raman scattering)effect due to its superior capability of Raman signal enhancement since its discovery in 1974.The bio-detection technology based on SERS effect has gradually become one of the most promising optical detection technologies,with many advantages such as high detection sensitivity,multi-dimensional parallel detection and non-destructive testing.In this paper,a method for detection of fusion protein BCR-ABL based on SERS technology is proposed,which can achieve qualitative and quantitative detection of the fusion protein.The method uses magnetic metal nanoparticle modified with anti-BCR antibody as SERS substrate and anti-ABL antibody modified silver-coated gold nanorods combined with 4-MBA as SERS immune-probe.The detection is achieved by “sandwich” immune-detection protocol.In unit of concentration of K-562,the dynamic detection range can reach 0.05%-10%,and the limit of detection is 0.05%.On the basis of the above,the level of BCR-ABL fusion protein in whole blood of clinical leukemia patients is tested.The experimental results show that the detection method can distinguish BCR-ABL positive blood samples from the negative one.
