Hyperlipoproteinemia is a disorder of dyslipidemia that usually shows elevated levels of serum total cholesterol(TC),triglyceride(TG)or low density lipoprotein(LDL-C),while the level of high density lipoprotein cholesterol(HDL-C)decreased,which seriously endangers human health and its incidence rate also increased year by year.At present,the synthetic lipid-lowering diet drugs sold in the market have been questioned by people because of side effects.Therefore,the research and development of safe,non-toxic and highly effective natural lipid-lowering weight loss products has become a research hotspot.Tea,as a natural beverage,has attracted much attention for its drinking history and health function.As a representative tea product of Hunan dark tea,Fuzhuan tea is favored by people for its unique quality flavor;but young people are no longer satisfied with the traditional drinking flavor.This research intends to develop an innovative tea product with no side effects,good taste,safety and functionality in order to enrich the form of Fuzhuan tea products and expand its sales market.Formula optimization,functional research and safety evaluation were carried out with raw materials such as sapphire tea,hawthorn,chamomile and mint.The research results are as follows:1.Formulation research and biochemical composition analysis of compound teaThe compound tea with Fuzhuan tea as the main material and homologous plant materials such as hawthorn,chamomile and orange peel as the auxiliary materials was tested on the basis of single factor.The results showed that the optimal formula and ratio are hawthorn 5.0g,dandelion root 5.0g,black tea 10.0g,mint leaf 6.0g,cumin 3.0g,dried tangerine peel 5.0g,chamomile 5.0g,which had bright soup and good taste.The content of flavonoids,tea polyphenols and amino acids was 4.83%,11.59% and 4.12%;the content of water extract was 37.56%.2.compound tea lipid-lowering effectThe hyperlipidemia mouse model was induced by high-fat diet.The mice in the normal group,the model group,the positive group and the high-,medium-,and low-dose groups of the tea were intervened to test the body weight and blood lipids.The results showed that:(1)The compound tea group could inhibit the weight gain of mice in different degrees;(2)The serum levels of TC,TG,LDL-C in the various dose groups of compound tea decreased,the level of HDL-C increased,and blood lipid level was effectively adjusted;(3)combined with the pathological section of liver tissue,it can be seen that the compound tea group can effectively improve liver tissue lesions,especially in the high-dose group of compound tea;(4)Compound tea could reduce SOD activity in mice liver and increase MDA activity,which relieves oxidative stress and improves free radical metabolism balance to some extent.3.Safety evaluation of compound teaThe acute toxicity test combined with the bacterial back mutation test(Ames test),the mammalian red blood cell micronucleus test,and the in vitro mammalian chromosome aberration test were used to evaluate the safety of the compound tea.The results showed that:(1)The maximum dose of the extract was administered to the mice once,and no death occurred after 7 days.It can be seen that the maximum tolerated dose was greater than 21.285g/kg.bw,which confirms that the compound tea is non-toxic;(2)Ames test,mammalian red blood cell micronucleus test,and in vitro mammalian chromosome aberration test results were all negative,indicating that the complex tea is not genotoxic,and long-term proper drinking is safe and reliable.
