With the improvement of people's living standards,environmental urbanization and excessive nutrition has led to the rise in lipid metabolism diseases.Diet-induced diseases have become a long-term research hotspot of scientists.According to the principle of “prevention treatment of disease”,it has become a global concern to actively seek safe and effective food to prevent and treat obesity and hyperlipidemia.Areca catechu and fuzhuan brick tea as a natural plant product which have significant effect of reducing obesity and hyperlipidemia.however,the synergistic effect has never reported.Therefore,In this study,fuzhuan brick tea water extract with different addition amounts of areca nut(5%,10%,20%)(AFT for short)was taken as the research object,and the fuzhuan tea group without adding areca nut was used as the control group.Mice were continuously gavage for 28 days under the condition of high-fat diet intervention,to explore fuzhuan tea water extract with different areca nut addition on therapy hyperlipidemia and adiposity.The relative abundance of microbes and changes in microbial community structure in mice feces were studied by high-throughput sequencing technology to further explore the relationship between AFT-obesity-microbes.The main conclusions described are as follows:Firstly,the functions of reducing obesity and hyperlipidemia of AFT were evaluated.Eight-week-old male kunming mice were selected as experimental animals.After a week of adjustable feeding,they were randomly divided into 5 groups(n=12)according to their body weight,namely the normal group(N),the control group(FT),the 5% AFT group(LAF),the 10% AFT group(MAF),and the 20% AFT group(HAF).The weight and food intake of mice were assessd every day during the experiment.At the end of the study,liver and epididymal adipose tissues were measured after which they were killed.The results showed that AFT had an inhibitory effect on the weight gain of mice and could increase the food intake.It was mainly found in 20% AFT group,but there was no significant dose relationship with the increase of the content of areca nut.AFT can effectively improve the phenomenon of liver cells increase and the accumulation of liver fat and epididymal adipose tissues caused by high-fat diet mice,HAF group is the closest to the normal group.Compared to the control group,liver cell shrinkage,The nucleus were arranged neatly,the cells size were uniform,the intercellular space was clear,and there were occasional fat vacuoles in the cytoplasm,with a low degree of steatosis.Secondly,to explore the effects of fuzhuan tea water extracts with different amounts of areca nut on biochemical and immune indexes of mice.The results showed that AFT could reduce the disorder of blood lipid and glucose metabolism,The contents of TC,TG,HDL-C and GLU in the 20% AFT group decreased by 2.35%,7.35%,4.97% and 9.58% respectively compared with the control group.5%,10% and 20% of the group could regulate lipid metabolic diseases induced by high-fat diet by reducing the level of bile acid and the content of free fatty acid,so as to promote the digestion of lipids and reduce the secretion of cholesterol.Results from immune index,The contents of IL-2?IL-4?IL-10 increased in 10% and 20% AFT group,while the TNF-? and IFN-? levels decreased significantly.When the amount of areca nut reach20%,the best effect was achieved in improving the inflammatory response induced by the highfat diet.Finally,The effects of fuzhuan tea water extracts with different amounts of areca nut on the faecal flora of mice were also investigated by high throughput sequencing technology.The results showed that AFT could significantly increased the relative abundance of Firmicutes,and decrease the relative abundance of Bacteroidetes.Compared with FT group,the indexes of Chao1,ACE,Shannon and Simpson in the 20% AFT group decreased,indicating that the higher content of areca nut could inhibit the microbial richness and diversity.The levels of Lactobacillus and Helicobacter in the 5%,10% and 20% groups were significantly higher than control group.Bifidobacterium in the 10% AFT group and Romboutsia in the 20% AFT group were significantly higher than the control group.In this paper,preliminary study on the correlation analysis of the blood index and genus was reported.Romboutsia and Lactobacillus were negatively correlated with TG and body weight,while Akkermansia and Bifidobacterium were significantly different from TC and HDL-C.The results showed that AFT could promote the growth of probiotics(such as Romboutsia,Lactobacillus and Akkermansia)and prevent or treat obesity and related metabolic disorders.In summary,fuzhuan tea water extracts with different amounts of areca nut had the effect of reducing blood lipid and glucose.AFT could effectively inhibit the weight growth of mice and then improved the phenomenon of liver cell enlargement and fat accumulation of liver and epididymis caused by high-fat diet,Meanwhile,AFT could cure lipid metabolism related diseases by regulating the diversity and structural changes of intestinal flora.However,the mechanism of the interaction between areca nut and fuzhuan brick tea need further study,which not only provides a theoretical basis for areca product processing,but also provides a reference for the future direction of areca industry.
