Objective:(1) To investigate the lipid-regulating effect of Compound Ni a-cuo Extract (CNAE). (2) To establish the best extractionprocess of CNAC. (3) To determine the molding process conditions of the CNAC.Method:(1)The rat hyperlipidemia model was set up by ig lipid emulsion. The rats were randomly divided:normal control group,model group,positive control group (1 mL·kg-1),SAE high dose group (12 g·kg-1) and low dose group (6.0 g·kg-1),CNAE high dose group (16 g·kg-1) and low dose group (8.0 g·kg-1). Except control group,other groups were given lipid emulsion and fed basal feed.The blood was collected from the aorta abdominalis of rats for measuring the serum TC,TG, LDL-C,HDL-C,MDA,SODand GSH-Px. (2) Using the orthogonal design experiments and single factor experiments,with the extract percent and the content of ferulic acid as the indexes,to confirm optimal conditions. (3) To investigate the effects of different matching excipients on the moisture absorption of extract powder; To research the optimum technique of granulation processes with orthogonal test method.Result:(1)CNAE could significantly decrease the serum contents of TC,TG, LDL-C,MDA in the experimental hyperlipidemic rats (P<0.01),and markedly increase the level of serum HDL-C,the SOD and GSH-Px activities of serum in the group of CNAE were much higher than those in hyperlipidema group.Also found, CNAE for reducing lipid effect better than the SAE. (2) The best extraction conditions include the ration of 1 (medicine herbs):10(water), backflow extraction 2 times with the water temperature of 95 centigrade and 1 hours every time. (3) The experimental results proved that extract powder absorptivity could effectively reduce with addition appropriate proportion of excipients; The optimum process was:extract powder,soluble starch and dextrin in proportion as 1:0.5:0.5 mixed with 70% ethanol.The granules were uniform and epigranular.Conclusion:(1) These results suggest that CNAE possess obvious lipid-regulating and antioxidative effects. Such characteristics will be of significance to prevent and/ or treat hyperlipidemia. (2) The optimized extraction conditions of CNAC is reasonable and feasible. (3)The optimum process is available for development and production of CNAC.
