Hawthorn pectin oligosaccharide(POS)is an oligosaccharide induced from the pectin in the fruit of hawthorn.It is an excellent water-soluble dietary fiber,which has the functions of enhancing the body's immunity,anti-oxidation,and anti-inflammatory.In this paper,POS was used as the test material,and mice fed with high-fat diets were used as animal test models.Using molecular nutrition techniques such as fluorescent quantitative PCR and enzyme-linked immunoassay,this paper studied the effects of POS on adiponectin in adipose tissue of high-fat diet mice(The effect and mechanism of ADPN)synthesis and lipid metabolism are as follows:(1)POS can inhibit the increase in body weight of high-fat mice,reduce serum TC,TG and FFA levels,and increase serum ADPN content.At the same time,POS can also significantly reduce the levels of inflammatory factors TNF-? and IL-6 in adipose tissue of high-fat food mice,and down-regulate the expression of CD68 gene.These results indicate that POS can improve abnormal lipid metabolism and inflammation in high-fat diet mice.(2)The analysis of adiponectin synthesis pathway-related regulatory factors in white adipose tissue showed that POS up-regulated ADPN gene and protein expression in adipose tissue,increased c AMP content in adipose tissue,and up-regulated AC,C / EBP?,RXR,PPAR? gene and protein expression.POS also up-regulated the expression of PKA and Rap1 genes and proteins,and up-regulated the expression of Src,ERK,and CREB genes and their phosphorylated proteins.These results indicate that POS promotes ADPN synthesis in adipose tissue and it can activate C/EBP?/PPAR? and PKA/ERK/CREB signaling pathways in adipose tissue.(3)Analysis of the effect of POS on lipid metabolism in white adipose tissue showed that POS could significantly increase the expression of ADPN receptor Adipo R1,AMPK and PPAR? genes in adipose tissue of high-fat diet mice.In addition,compared with the high-fat model control(HFC),POS intervention up-regulated the gene expression levels of triglyceride catabolism-related enzymes ATGL,HSL,and fatty acid oxidation-related enzymes CPT-1 and ACO in adipose tissue;and down-regulated adipose tissue.Medium fatty acid synthesis-related enzymes ACC,FAS and gene expression levels of related enzymes SCD1 and DGAT1 that regulate the synthesis and deposition of fat droplets in tissues.These results show that POS can promote ADPN-mediated Adipo R1 / AMPK / PPAR? and Adipo R1 / AMPK / ACC signaling pathways in adipose tissue,thereby promoting the oxidation of fatty acids and the decomposition of triglycerides in adipose tissue,and inhibiting fatty acids and fatty acids in adipose tissue.Synthesis of triglycerides.The above results show that POS can be used as a new food ingredient in the development of foods that improve dietary obesity and high blood lipids.
