In recent years,food safety issues have attracted more attention as a major livelihood issue.As a strong carcinogen,benzopyrene(BaP)is an important test indicator for the quality of oils and fats.It is imperative to develop a rapid test method for BaP.Surface-Enhanced Raman Scattering(SERS)has the advantages of strong specificity,high sensitivity,simple operation and low cost,and is widely used in the field of rapid food safety detection.The development of a rapid BaP detection method based on the SERS method is of great practical significance.This paper successfully constructed a rapid and sensitive detection method for BaP in animal and vegetable oils based on SERS method.The main research contents are as follows:First,we synthesized gold-plated silicon wafers,colloidal gold and colloidal silver sol,and use UV-visible spectrophotometer,Transmission electron microscope and other equipment to investigate the physical and chemical properties of the three substrate materials.It is found that the gold-plated silicon wafers have good mechanical properties and stability.The colloidal gold sol has uniform particle size and stable state with high performance,and the colloidal silver sol has uniform particle size and diverse forms.Using the BaP standard solution of a specific concentration to initially verify the enhancement effect of the three base materials,the results show that the enhancement effect of colloidal gold on BaP is the most significant,followed by colloidal silver and gold-plated silicon wafers.Since colloidal gold has the advantages of good stability and easy storage,it is especially suitable for on-site rapid detection for BaP.Then,we optimized the pre-treatment method for rapid detection of BaP.Respectively optimize the coagulation reagent,extraction reagent and solid phase extraction packing.After comparing various reagents,the results show that sodium chloride has the best coagulation effect,acetonitrile has the highest extraction efficiency,neutral alumina has a high recovery rate of BaP with a good impurity removal effect.Finally,based on the mentioned SERS material and optimized pre-treatment methods,the on-site rapid qualitative and quantitative detection of BaP in animal and vegetable oils was carried out.It was found that the established detection method has excellent technical indicators and performance:the detection limit can reach 5?g/L,The limit of quantification is 15?g/L,the linear range is between 0-100?g/L,and the correlation linear coefficient can reach 0.99,which ensures the accuracy of BaP detection;investigated the effects of other toxic and hazardous substances on BaP.The interference situation of the detection showed that the four substances of Rhodamine B,Rhodamine 6G,Sudan Red 1 and Sudan Red 2 did not interfere with the detection of BaP,which proved that the method has good anti-interference performance;The chili oil was tested on actual samples,and the recovery rate was between 76.9%and 105.8%;and the parallel comparison test with the national standard method showed good consistency,indicating that the method established in this paper has better detection performance.The method established in this article is compared with the traditional BaP detection method,the rapid detection method established in this article has the advantages of simple operation and low cost.It is suitable for rapid detection and use in actual process and will improve the ability of relevant units and enterprises in the food safety field to supervise food safety risk factors.
