Diabetes is a chronic disease caused by endocrine disorders,which main feature is hyperglycosemia.With the development of society,diabetes has become one of the killers that endanger people's health.At present,there is no specific medicine for curing diabetes.The main function of current medicine is to control the patients' blood glucose levels.Long-term using will cause drug dependence and be harm to the body.Eggplant is a kind of common food,and eggplant contains a variety of flavonoids,anthocyanins,alkaloids and other substances,which have a certain hypoglycemic effect.This article uses purpleblack eggplant skin as raw material to study the components of eggplant skin extraction and its hypoglycemic effect,and prepare the lozenge which suitable for diabetes.The study provides a theoretical reference for the further development and utilization of biologically active substances in eggplant skin.This research mainly includes the following parts:1.Screening of eggplant skin and drying method.UPLC MS/MS was used to analyze the content of phenols and flavonoids in purple-red eggplant skin and purpleblack eggplant skin.The study showed that the phenolic compounds in purple-black eggplant skin were significantly higher than those in purple-red eggplant skin.The skin is the raw material;using LC-ESI-Q-TOF-MS to analyze hot-air drying and freeze-dried eggplant skin extracts,the results show that hot-air drying can retain 11 kinds of active ingredients,and freeze-drying can only retain 9 compounds,so hot-air drying is selected.2.Study on the extraction and functional characteristics of eggplant peel extract.The eggplant skin was extracted with water as the extractant.On the basis of the single factor experiment,the orthogonal experiment was used to optimize the extraction process.The study showed that the ratio of material to liquid was 1:50 g/m L,the extraction time was 50 min,and the ultrasonic power was 540 W.,The extraction temperature is 50 ?,and the experiment is repeated 3 times under this condition.The average yield of the extract is 43.21%.The effect of eggplant peel extract(EPE)on ?-glucosidase activity was studied.The results showed that EPE has a certain inhibitory effect on ?-glucosidase activity,and the inhibitory effect increases with the increase of extract concentration.3.Preparation of eggplant skin lozenges.Taking sensory evaluation as the evaluation index,the final optimized formula of eggplant skin lozenges was determined to be: EPE 30%,microcrystalline cellulose 15%,xylitol 16%,inulin 12%,mannitol 12%,magnesium stearate 0.5 %,polydextrose 13%,citric acid 2.5%.The obtained lozenges have a smooth surface without cracks and a unique fragrance of eggplant.The weight difference of the lozenge is 0.253%?4.004%(<5%),the disintegration time of the tablet is about 10 min,the hardness of the tablet is 30 N?40 N;the friability is 0.4886%,and the weight loss rate is less than 1%.It meets the inspection requirements of the Chinese Pharmacopoeia for tablets.4.Preliminary study on the hypoglycemic effect of eggshell buccal tablets.Taking acarbose as a positive control,the hypoglycemic study was conducted on diabetic mice,and body weight,fasting blood glucose,and glucose tolerance were used as evaluation indicators.Compared with the model group,the fasting blood glucose level and glucose tolerance of diabetic mice in the high-dose group of eggplant buccal tablets were significantly decreased(P<0.05),indicating that eggplant buccal tablets can effectively reduce the blood sugar of diabetic mice.
