Generally speaking,most drugs like vaccines can be delivered through hypodermic injection for providing a low-cost,rapid and direct way to deliver almost any type of molecule into the human body.But hypodermic needle brings many pains to patients and difficult to use by themselves,only people who receives professional training can use hypodermic needle.What’s worse,risk of injection and difficulty in obtaining sustained drug delivery are also a big problem we met especially in developing countries.Oral administration solves the problems generated by hypodermic injection and has the specific merits that oral drug delivery systems offer patient more compliance,convenient in transdermal delivery,rich blood supply due to the large surface area,low cost and easy to release drug in stomach and intestines.However,some macromolecular drugs like DNA,protein and vaccines cannot deliver through oral due to drug degradation in the gastrointestinal tract and first liver-pass effect,which cause the low drug absorption efficiency.To overcome the limitations of hypodermic injection and oral delivery,transdermal drug delivery system have caught most public scientists’ eyes.In the past few decades,among kinds of drug-loading microneedles,rapidly separating MNs attract human’s more attention on its superiority of enough mechanical strength to deliver drug into skin due to PLA solid microneedles,which can rapidly separate from the upper segment to release drugs.Compared with the widely used dissolving microneedles,rapidly separating microneedles overcome the inferior mechanical properties of dissolving microneedles.Gradually,we must choose the appropriate angle and direction to research it.Microneedles are used to serve for people and can improve our life.And how to use MNs,when to deliver drugs and how to store MNs are very crucial for MN’s fabrication and application.We choose RSMN for our study to give strong mechanical strength at high humidity environment,which could give more information in further.In addition,we have design and prepare coating microneedles with a novel method,which solved the two problems(low and inhomogenous drug loading).
