Raman spectroscopy is convenient,fast,low cost,non-destructive,pollution-free,known as the"fingerprint spectrum."In recent years,Raman spectroscopy detection technology has broad application prospects in the fields of petrochemical and basic organic chemical,polymer chemical,pharmaceutical and clinical medicine,bio-chemical,environmental science,drug identification,gem identification and so on.In this paper,the optical design of the portable optical fiber spectrometer is mainly discussed.According to the aberrations of the Czerny-Turner cross-structured optical structure,theoretical analysis of the primary elimination of coma and astigmatism is made,and the optical path is simulated and optimized using Zemax software.And the mechanical structure of the prototype and the related regulating device are designed.On this basis,a prototype of near-infrared Raman spectrometer based on Czerny-Turner cross-section is designed with 1064nm laser as the excitation source.The wavelength range of the spectrometer is 1080nm-1580nm,the spectral resolution is about 1.8nm.The dimensions are 142×114×68 mm~3.In this paper,the R-spec of Raman spectrum data acquisition software is designed and developed.In addition to the basic function of spectral data acquisition,the acquisition software also includes the functions of spectral peak searching,wavelength calibration and so on.In addition,the noise,fluorescence background,sample difference and the extraction of characteristic variables in Raman spectroscopy are discussed in detail.The PCA-LDA model was used to rapidly identify and classify the different series of hydroxypropyl methyl cellulose(HPMC),a widely used pharmaceutical adjuvant,which shows the prospect of Raman spectroscopy in field of drug production and manufacture.
