In recent years,the traditional microscope can realize super resolution imaging by combining with transparent dielectric microspheres.This new super resolution imaging technique is simple to operate and easy to implement,which provides a new method for the large-scale real-time super-resolution imaging of sub-diffraction limit samples and biological tissues.This paper studies and constructs a super resolution imaging system based on integrated microsphere objective lens.After analyzing the imaging mechanism of encapsulated microspheres with different parameters,the electronically controlled microsphere component is developed,which enables traditional microscopes to achieve super resolution imaging.In order to improve the practicability and integration of the device and analyze the imaging parameters of microspheres,an integrated microsphere objective lens is designed and the corresponding superresolution imaging setup is built.Integrated microsphere objective lens can realize realtime damage-free super-resolution imaging of the controllable region of the chip lattice feature and the blu-ray disc fringe feature at different working distances successfully,which greatly improves the imaging performance of the common objective lens and the applicability of the microsphere super-resolution technology.The main contents and innovations of this paper are as follows:1.The study of mechanism of super resolution imaging based on microspheres was carried out.The mathematical model is established to analyze the focusing optical characteristics of microspheres,which studies the effect laws of the depth of immersion,the refractive index and diameter of the encapsulated microsphere on the imaging performance of the microsphere.Furthermore,the selection criteria of encapsulated microsphere parameters in different applications are discussed.2.According to the traditional microscope,the electronically controlled microsphere component is designed to achieve the super-resolution imaging of controllable region under white light.The component can be used without changing the structure of traditional microscope,which can control the distance among the microsphere,objective lens and the sample by stepping motor.This component greatly improves the performance of the traditional microscope.3.The super resolution microscopic imaging system based on integrated microsphere objective lens is designed and established to achieve real-time undamage super-resolution microscopic imaging of controllable region.The self-made metal sleeves are fabricated to integrate the microspheres with the ordinary objective lens,which greatly improves the imaging performance of the objective lens.The resolution of the ordinary microscopic objective lens(40×,NA0.6)can be improved by 4.78 times,and the sample characteristics of 100 nm can be resolved.This design greatly improves the versatility of microspheres super-resolution technology.4.The side-view imaging feedback system is established,the camera and the adjustable focus lens and the electric displacement platform,in which,can control the distance among the microsphere,objective lens and the sample precisely to adjust the imaging parameters to achieve super-resolution imaging.In addition,the impact of working distance on magnification,image contrast and other imaging parameters in microsphere imaging is studied.5.The integrated microsphere objective lens is successfully manufactured by the self-developed method of fabrication,the equipment or components such as self-made spin coater,heating platform,electric nanometer displacement table and the side-view imaging system are used.These components can be used to make integrated microsphere objective lens with different diameters microspheres easily,quickly and cost effectively.The microsphere objective lens,making in this way,is robust,easy-touse and reusable.
