Scutellaria baicalensis,belonging to the family Labiate,is a perennial herb.The bioactive components of Scutellaria have been confirmed to be flavones,which are the molecular and material basis of pharmacological activity.They are mainly produced by secondary metabolites of plant,and the phenylpropanoid pathway is considered an important regulation point between primary and secondary metabolism.Biological metabolism of flavonoids is an important branch of phenylpropanoid metabolic pathways.The content of the total flavonoids were assayed during the different growth period of S.baicalensis from Changbai Mountain area.and it showed different flavonoids accumulation mechanisms in the adventitious roots,the callus,annual and biennial roots of S.baicalensis.Therefore,the optimal harvest time is June for annual S.baicalensis,and September for biennial S.baicalensis.The 40d for adventitious root,the 28 d for callus after inoculating.In order to explore the relationship between the accumulation of total flavonoids and the transcriptional expression of the genes of phenylpropanoid metabolic pathway during the growth and development of S.baicalensis.Therefore,the full length cDNA of SbPAL(SbPAL1,SbPAL2,SbPAL3),SbC4H and Sb4CL sequence have been cloned.The results showed that the length of three PAL cDNA sequences are different,2532 bp?2275 bp?2485 bp respectively,encoding 708?670?704 amino acids respectively.Similarly,C4H cDNA is 1724 bp encoding 507 amino acids;The 4CL cDNA is 1858bp encoding 549 amino acids.And bioinformatics analysis revealed SbPAL and Sb4CL are both hydrophilic proteins,and they localized in the cytoplasmic matrix,SbC4H had signal peptide.Semi-quantitative expression analysis showed that it is a very complicated process in the accumulation of total flavonoids and the genes expression inphenylpropanoid metabolic pathway.SbPAL?SbC4H?Sb4CL sustained expression in every growth period.These genes have different expression patterns in the annual and biennial S.baicalensis,and it's also different in the callus and adventitious roots.
