Brazilian rubber tree(Hevea brasiliensis)belongs to the Euphorbiaceae,Hevea plants,and also called Hevea and rubber trees.This study was to investigate the rubber of 0.5y cultured seedling physiolodical and biochemical changes in the growth process,and understood the growth dynamic in seedling rubber tree.Provide the certain theoretical for fast-growing traits of rubber tree genetic improvement.This study selected the Reyan 7-33-97 0.5-year-old seedlings of hevea brasiliensis stirpe's four different developmental stages of wood as a material,Study four development stages of that is light green bark wood(LGX),green bark wood(GX),half a Corky bark wood(SSX)and Corky bark wood(SX).Rubber stem observed with scanning electron microscope microstructure of wood of different development stages,and the stem wood of hevea brasiliensis at different developmental stages of lignin,cellulose,hemicellulose and dynamic changes of soluble sugar content,using 2-D SDS-PAGE to tap rubber stem wood protein of differentially expressed in different development stages,and use LC/MS technology to reveal the different developmental stages of metabolic in stem wood.Results of this study as follows:1,Under electron microscope view of the xylem cell wall as growth gradually thickens.The amyloplasts in xylem cell was gradually increase.The amyloplasts's shape have changed from round to drop-shaped.And discover 3 types of tube in rubber seedling:central line catheters,catheter and trapezoidal thread.catheters2,Measured the content of lignin,cellulose and hemicellulose were gradually increased along with the growth and development.Trend along with the growth of soluble sugar in general is reduced.3,Protein extraction of the xylem materials by TCA-acetone-phenol method,the protein production of LGX period was up to 8.36 mg/g,GX period was 7.44 mg/g,SSX was 4.16 mg/g and SX period was 3.14mg/g,respectively.the protein content was decreased gradually with the growth of seedling.The extracted proteins were staining after SDS-PAGE electrophoresis and two-dimensional electrophoresis,and the bands and cbservey clearly of bands in mini gel and a high quality 2-D maps.Find the different protein from four stage by PDquest software and to mass spectrometry,eliminating false positive results,received a total of 24 proteins,identified by the database search function of these proteins.PPR protein families took part in plant chloroplasts and mitochondrial RNA processing,cytoplasmic male sterility of recovery,is also involved in the formation and regulation of plant growth and development of embryos.MCM proteins are a small chromosome repair protein,plays a role in the regulation of DNA replication.4,The study of metabolism in the seedling tubber,identification 64 differences metabolism in positive,are ion mode and 26 differences metabolism in negative mode.anion mode.Removed non-plant of metabolites got 39 differences metabolism finally.Online database of metabolism way analysis,which of 15 of metabolism real determine has 22 a metabolism way.
