| In recent years,it becomes more and more popular to resequence different species g enome with the rapid development of sequencing technology.Through the analysis of a certain species resequencing data of different varieties,the selected area can be found in the entire process of domestication.With the analysis of genetic selection region,we will have a lot of traits associated with a specific candidate genes,and identify the genes function in the groups.After the early comparative analysis of resequencing genome of Tong Cheng Pig with European wild boar and domestic pig,we found there were 196 select sweep region from autosomes and X-chromosome.We took 570 protein-coding genes in these areas for Gene Ontology analysis,these genes enriched in 34 different functional clustering items,including 2 items behavior related.Based on the candidate genes’information of select sweep region related to the pig’s length,this study focused on the candidate genes related to the number of ribs.This study carried out the measure of aggressive behavior and the number of ribs,the filter of candidate SNPs,genotyping and analysis associated with the trait.The main findings are as follows:1.Based on the selected sweep region of our group-wide gene resequencing analysis to identify the candidate genes about the aggressive behavior.Our team found the select sweep region about behavior in the process of domestication though the group-wide gene resequencing analysis,the selected region related to behavior was found.Four nervous system-related candidate genes(PPFIA3,YWHAZ,SERPINI1 and GLRB)were screened out.By Bulked Segregant Sequencing,we found there are two missense mutations in the fourth and nineth exons in the Large White pigs;We observed the Large White pigs by using vedio and manual observation for 24 hours after the gregariousness and gave the skin lesion score after 24 hours.We measure the aggressive behavior by counting the numbers of individual battle and skin lesion score;By constructing the gene-trait association analysis model,the g.50073.C>G of SERPINI1 has association with aggressive behavior,and the genotype of GG are significantly lower than the genotype of CC in the number of battle,front skin lesion,middle skin lesion and the whole skin lesion.It shows that this SNP can be used as the marker loci of aggressive behavior.2.Identification of candidate genes about the number of ribs in the "E Tong Liang Tou Wu"groups based on the select sweep regionBased on the report of select sweep region about the body length of pigs,we choose VRTN、NR6A1、-PLAG1、LCORL as the candidate genes;We slaughtered 398 pigs and accurately recorded the number of ribs;By constructing the gene-trait association analysis model,we detected the 291bp insertion homozygotes of VRTN has 0.9 more pair of ribs and 1.7 more pair of ribs than heterozygous and deletion homozygotes;The results showed that the TT genotype of NR6A1 has 0.5 more pairs of ribs than TC genotype and 0.9 more pairs of ribs than CC genotype;The GG genotype of PLAG1 has 0.6 more pairs of ribs than GT genotype and 0.9 more pairs of ribs than TT genotype;VRTN and NR6A1 have the significant gene interaction effects.The individual with 291bp insertion homozygotes of VRTN and TT genotype of NR6A1 has the most number of ribs.So NR6A1、PLAG1 are significantly associated with the number of ribs and can be the genetic marker of the number of ribs in the"E Tong Liang Tou Wu"group.In this study,we took association analysis between aggressive behavior and the number of ribs with candidate genes and found SNPs which can be used for effective marker-assisted selection.It also provided a reference for the study of gene function in the future. |
